貴州地方山羊FSHR基因與繁殖性狀的相關(guān)性研究

【摘要】:促卵泡素受體(follicle stimulating hormone receptor,FSHR)參與卵泡的生長(zhǎng)發(fā)育和分化成熟,與動(dòng)物的繁殖性狀存在一定的關(guān)聯(lián)性。為了探究FSHR基因多態(tài)性及其m RNA的表達(dá)量與黔北麻羊(Capra hircus)繁殖性狀之間的關(guān)系,本研究選取高繁殖力的黔北麻羊和低繁殖力的貴州黑山羊(Capra hircus)為實(shí)驗(yàn)對(duì)象,采用聚合酶鏈?zhǔn)椒磻?yīng)-單鏈構(gòu)象多態(tài)(polymerase chain reaction-single strand conformation polymorphism,PCR-SSCP)和PCR技術(shù)對(duì)2個(gè)品種FSHR基因第1和10外顯子的多態(tài)性進(jìn)行檢測(cè),并分析突變位點(diǎn)與產(chǎn)羔數(shù)之間的相關(guān)性。結(jié)果顯示,2個(gè)實(shí)驗(yàn)群體在第1和10外顯子各存在1個(gè)SNP位點(diǎn),即C126T和C1246A。在黔北麻羊中,BB型個(gè)體的產(chǎn)羔數(shù)顯著高于AA型和AB型(P0.05);在貴州黑山羊中,2個(gè)SNPs位點(diǎn)不同基因型個(gè)體之間的產(chǎn)羔數(shù)均差異不顯著(P0.05)。此外,q RT-PCR結(jié)果顯示,FSHR基因在BB型黔北麻羊各組織的表達(dá)量均高于AA型,且在BB型黔北麻羊發(fā)情間期下丘腦組織和發(fā)情期卵巢組織的表達(dá)量極顯著高于AA型黔北麻羊(P0.01),在發(fā)情間期和發(fā)情期的子宮組織中FSHR基因表達(dá)量在兩種基因型的個(gè)體間也達(dá)到顯著差異水平(P0.05)。結(jié)果提示,FSHR基因?qū)τ谡{(diào)控黔北麻羊的多羔性狀具有重要作用。研究結(jié)果為從分子水平探索影響黔北麻羊繁殖性能的候選基因,以及建立高繁殖力的貴州地方山羊核心群提供基礎(chǔ)資料。
[Abstract]:The follicle stimulating hormone receptor (follicle stimulating hormone receptor, FSHR) participates in the growth and differentiation of follicles and has a certain correlation with the reproductive traits of the animals. In order to explore the relationship between the polymorphism of the FSHR gene and the expression of M RNA and the reproductive traits of the northern Guizhou sheep (Capra hircus), this study selects the high fecundity of Guizhou. North Ma sheep and low fecundity Guizhou black goat (Capra hircus) were tested by polymerase chain reaction single strand conformation polymorphism (polymerase chain reaction-single strand conformation polymorphism, PCR-SSCP) and PCR techniques to detect the polymorphism of first and tenth exons of 2 FSHR genes, and the mutation sites and production were analyzed. The correlation between the number of lambs showed that there were 1 SNP loci in the first and tenth exons of the 2 experimental groups, that is, C126T and C1246A. in the northern Guizhou sheep, the number of lambing of BB type individuals was significantly higher than that of AA and AB (P0.05). In Guizhou black goats, there was no significant difference in the number of lambs among the 2 SNPs loci in Guizhou black goats. Moreover, The results of Q RT-PCR showed that the expression of FSHR gene in all the tissues of BB sheep in northern Guizhou was higher than that of the AA type, and the expression of the hypothalamus and the oestrus of the oestrus was significantly higher than that of the AA type Qian North Ma sheep (P0.01). The expression of the FSHR gene in the uterine tissues of the estrus and estrus period was two genotypes in the oestrus and estrus period. The results suggest that the FSHR gene plays an important role in the regulation of the multi lamb traits in the North Guizhou Ma sheep. The results provide the basic information for exploring the candidate genes affecting the reproductive performance of the North Guizhou sheep from the molecular level, as well as the establishment of the high fecundity Guizhou local goat core group.
