棉花GhbHLH基因的克隆與表達(dá)分析

【摘要】:bHLH(basic helix-loop-helix,bHLH)家族基因在植物生長(zhǎng)發(fā)育和非生物脅迫應(yīng)答中發(fā)揮著重要的作用。本研究從前期構(gòu)建的棉花cDNA文庫(kù)中發(fā)掘得到一個(gè)新的GhbHLH基因,該基因開(kāi)放閱讀框全長(zhǎng)600 bp,編碼199個(gè)氨基酸,序列分析表明,該基因具有典型的helix-loop-helix結(jié)構(gòu)域。熒光定量PCR表明,GhbHLH在棉花根、莖、葉和纖維中均有表達(dá),并且能夠受高鹽、PEG、低溫、ABA和Me JA等的誘導(dǎo)表達(dá),亞細(xì)胞定位結(jié)果表明,GhbHLH在細(xì)胞核內(nèi)表達(dá)。推測(cè)該基因可能作為調(diào)控因子在棉花非生物脅迫應(yīng)答中發(fā)揮作用。
[Abstract]:The genes of bHLH(basic helix-loop-helixhh family play an important role in plant growth and development and abiotic stress response. In this study, a new GhbHLH gene was obtained from the cotton cDNA library constructed in the previous period. The open reading frame of the gene was 600bp in length, encoding 199 amino acids. Sequence analysis showed that the gene had a typical helix-loop-helix domain. Fluorescence quantitative PCR showed that GhbHLH was expressed in cotton roots, stems, leaves and fibers, and could be induced by PEG, low temperature ABA and me JA. The subcellular localization results showed that GhbHLH was expressed in the nucleus. It is speculated that this gene may play a role as a regulatory factor in cotton abiotic stress response.
