PTEN基因及miRNA-21在新疆漢族2型糖尿病中的表達(dá)及其相關(guān)性研究

【摘要】:目的:運(yùn)用實(shí)時(shí)熒光定量PCR(q RT-PCR)技術(shù)檢測(cè)PTEN基因及mi RNA-21的表達(dá)水平,觀察PTEN及mi RNA-21表達(dá)水平在新疆漢族2型糖尿病(T2DM)患者與糖耐量正常(NGT)者中的差異,探討PTEN基因及mi RNA-21在新疆漢族T2DM中的表達(dá)及其相關(guān)性。方法:選取新疆漢族T2DM患者和NGT者各50例分別作為T(mén)2DM組和NGT組;測(cè)量并記錄一般資料,包括:性別、年齡、身高、體重、腰圍、臀圍、血壓等,計(jì)算體質(zhì)指數(shù)(BMI)、腰臀比(WHR);檢測(cè)空腹血糖(FPG)、餐后2小時(shí)血糖(2h PG)、空腹胰島素(FINS)、空腹C肽、糖化血紅蛋白(Hb A1c)、甘油三酯(TG)、總膽固醇(TC)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)等生化指標(biāo),計(jì)算胰島素抵抗指數(shù)(HOMA-IR);應(yīng)用q RT-PCR技術(shù)檢測(cè)兩組PTEN基因及mi RNA-21的表達(dá)水平;運(yùn)用SPSS 20.0軟件對(duì)數(shù)據(jù)進(jìn)行統(tǒng)計(jì)學(xué)分析。結(jié)果:1、新疆漢族T2DM組BMI、WHR較NGT組高,其差異有統(tǒng)計(jì)學(xué)意義(P0.05);T2DM組性別、年齡、SBP、DBP較NGT組差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。2、新疆漢族T2DM組FPG、2h PG、Hb A1c、ln(HOMA-IR)、TG、TC及LDL-C均高于NGT組,FINS、空腹C肽及HDL-C均低于NGT組,差異均有統(tǒng)計(jì)學(xué)意義(P0.05)。3、PTEN在新疆漢族T2DM組的表達(dá)水平是NGT組的2.18倍。4、mi RNA-21在新疆漢族T2DM組的表達(dá)水平是NGT組的2.12倍。5、在新疆漢族T2DM組中,PTEN基因與mi RNA-21的表達(dá)無(wú)明顯相關(guān)性(r=-0.078,P=0.592)。結(jié)論:1.PTEN基因和mi RNA-21在T2DM中的高表達(dá)可能參與了T2DM的發(fā)生發(fā)展過(guò)程。2.在新疆漢族T2DM中,PTEN基因與mi RNA-21的表達(dá)可能無(wú)關(guān)。
[Abstract]:Objective: to detect the expression of PTEN gene and mi RNA-21 by real-time fluorescence quantitative PCR(q RT-PCR, and to observe the difference of PTEN and mi RNA-21 expression in patients with type 2 diabetes mellitus (T2DM) in Xinjiang Han nationality and those with normal glucose tolerance (GTT). To investigate the expression and correlation of PTEN gene and mi RNA-21 in T2DM of Xinjiang Han nationality. Methods: 50 patients with T2DM and 50 patients with NGT in Xinjiang Han nationality were selected as T2DM group and NGT group respectively, and general data were measured and recorded, including gender, age, height, weight, waist circumference, hip circumference, blood pressure, etc. To calculate body mass index (BMI), waist-to-hip ratio (WHR), fasting blood glucose (FPGN), 2 hours postprandial blood glucose (FG), fasting insulin (FINSN), fasting C-peptide, glycosylated hemoglobin (HbA1cN), triglyceride (TGG), total cholesterol (TC), high density lipoprotein (HDL-C), low density lipoprotein (LDL-C), and so on. The expression levels of PTEN gene and mi RNA-21 in two groups were detected by Q RT-PCR technique, and the data were analyzed by SPSS 20.0 software. Results BMIP WHR in Xinjiang Han T2DM group was higher than that in NGT group, the difference was statistically significant (P 0.05). There was no significant difference in age (P 0.05). There was no significant difference between NGT group (P 0.05) and age group (P < 0.05). The TC and LDL-C of T2DM group were significantly higher than those of NGT group (P < 0.05). The fasting C-peptide and HDL-C were lower in T2DM group than in NGT group. There were significant differences in the expression level of P0.05PTEN in T2DM group of Xinjiang Han nationality and 2.18 times of that in T2DM group of NGT group. The expression level of RNA-21 in T2DM group of Xinjiang Han nationality was 2.12 times higher than that in T2DM group of NGT group. There was no significant correlation between the expression of PTEN gene and the expression of mi RNA-21 in T2DM group of Xinjiang Han nationality. Conclusion: 1. The high expression of PTEN gene and mi RNA-21 in T2DM may be involved in the genesis and development of T2DM. The expression of mi RNA-21 may not be related to the expression of PTEN gene in T2DM of Han nationality in Xinjiang.
