小鼠iPS細(xì)胞體外自發(fā)性分化及神經(jīng)定向分化中基因表達(dá)譜的研究

【摘要】:目的:研究小鼠誘導(dǎo)多能干細(xì)胞(i PS細(xì)胞)在體外自發(fā)性分化和神經(jīng)定向分化過(guò)程中的基因表達(dá)譜的特性。方法:在體外將i PS細(xì)胞分別進(jìn)行形成類胚胎小體后自發(fā)性分化及成骨蛋白抑制劑Noggin誘導(dǎo)神經(jīng)定向分化后,通過(guò)實(shí)時(shí)熒光定量PCR(q PCR)測(cè)定在分化過(guò)程中i PS細(xì)胞分化相關(guān)基因表達(dá)的變化。結(jié)果:i PS細(xì)胞形成類胚胎小體后胚胎外胚層標(biāo)志基因(GFAP,Map2和Tu J1)及內(nèi)胚層標(biāo)記基因(Foxa2,GATA4和Sox17)的表達(dá)隨分化而迅速增加,但中胚層標(biāo)記基因(Bmp4,BRA,FGF5)表達(dá)水平變化不明顯。在神經(jīng)定向分化過(guò)程中,i PS細(xì)胞的神經(jīng)標(biāo)志物基因(Map2,Neu N,Tu J1和Sox1)及線粒體相關(guān)基因(12s,ND3,ND5,ND6、Cytb和Cox1)的表達(dá)都逐漸增加,且兩者具有相同的增長(zhǎng)趨勢(shì)。結(jié)論:在自發(fā)性分化過(guò)程中小鼠i PS細(xì)胞具有自發(fā)向外胚層和內(nèi)胚層分化的趨勢(shì);在神經(jīng)定向分化過(guò)程中,i PS細(xì)胞的線粒體相關(guān)基因表達(dá)隨著分化表達(dá)逐漸增加,并與神經(jīng)細(xì)胞標(biāo)記基因具有正相關(guān)性,表明線粒體的功能在i PS細(xì)胞神經(jīng)定向分化中發(fā)揮重要作用。
[Abstract]:Aim: to study the characteristics of gene expression profile of mouse induced pluripotent stem cell / PS cells during spontaneous and neuronal differentiation in vitro. Methods: in vitro, I PS cells were divided into embryoid bodies after spontaneous differentiation and osteogenic protein inhibitor Noggin induced neuronal directional differentiation. The expression of differentiation related genes in iPS cells was measured by real-time fluorescence quantitative PCR(q. Results the expression of embryonic ectodermal marker genes (GFAPMap2 and Tu J1) and endodermal marker genes (Foxa2 (GATA4) and Sox17) increased rapidly with differentiation, but the expression level of mesodermal marker gene (Bmp4, BRAFGF5) did not change significantly. During the process of neuronal directional differentiation, the expression of neuronal marker gene (map2Neu Nu Tu J1 and Sox1) and mitochondrial associated gene (12sNd3Nd5Nd5Nd6, Cytb and Cox1) in PS cells increased gradually, and both of them had the same increasing trend. Conclusion: the spontaneous differentiation of mouse iPS cells into ectoderm and endoderm during spontaneous differentiation and the expression of mitochondria related genes in iPS cells increased with the differentiation. There is a positive correlation between mitochondria and neuronal marker gene, which indicates that mitochondrial function plays an important role in the neuronal differentiation of iPS cells.
