醌型二氫生物喋呤還原酶基因表達(dá)變化對高糖環(huán)境下NRK-52E細(xì)胞葡萄糖調(diào)節(jié)蛋白75表達(dá)的影響

【摘要】:目的探討醌型二氫生物喋呤還原酶(QDPR)基因?qū)Ω咛黔h(huán)境下腎小管上皮細(xì)胞系NRK-52E葡萄糖調(diào)節(jié)蛋白75(GRP75)的影響及其在DN中的可能作用機(jī)制。方法構(gòu)建過表達(dá)及敲低QDPR基因的NRK-52E模型,分別予5.4 mmol/L和30 mmol/L葡萄糖培養(yǎng)基培養(yǎng),Western blot檢測GRP75在OLETF大鼠腎皮質(zhì)及細(xì)胞模型各組的表達(dá)水平,流式細(xì)胞術(shù)檢測細(xì)胞周期。結(jié)果與LETO鼠比較,OLETF大鼠腎皮質(zhì)GRP75含量降低[(1.53±0.27)vs(0.79±0.26),P0.01];與NRK-52E組比較,NRK-52E高糖組的GRP75含量降低[(0.62±0.03)vs(0.46±0.06)]、G0/G1期細(xì)胞增多[(39.80±1.31)%vs(50.35±0.33)%]、S期細(xì)胞減少[(48.55±1.85)%vs(37.17±0.10)%](P0.05);過表達(dá)QDPR使高糖環(huán)境下GRP75含量降低[(0.85±0.10)vs(0.45±0.16),P0.05]、G0/G1期細(xì)胞增多[(43.73±0.48)%vs(61.87±0.17)%,P0.01],S期細(xì)胞減少[(42.42±0.66)%vs(25.29±0.12)%,P0.01];敲低QDPR不影響GRP75表達(dá)量與細(xì)胞周期。結(jié)論 GRP75在DN模型中含量減少,QDPR基因可能通過GRP75及細(xì)胞周期影響DN發(fā)生發(fā)展。
[Abstract]:Objective to investigate the effect of quinone dihydrotrexate reductase (QDP) gene on renal tubular epithelial cell line NRK-52E glucose regulatory protein 75 GRP 75 and its possible mechanism in DN. Methods the NRK-52E model of overexpression and knockout of QDPR gene was constructed. The expression of GRP75 in the renal cortex and cell model of OLETF rats was detected by Western blot in 5.4 mmol/L and 30 mmol/L glucose culture medium, respectively.
