刺芹側(cè)耳多肽對染鉛大鼠腦海馬組織形態(tài)及海馬組織中camk2a、erk2和homer1基因mRNA表達(dá)量的影響

【摘要】:為研究刺芹側(cè)耳(Pleurotus eryngii)多肽對染鉛大鼠腦組織損傷的影響,以雄性SD大鼠為研究對象,隨機(jī)分為正常對照組(基礎(chǔ)飼料,蒸餾水)、染鉛模型組(基礎(chǔ)飼料,0.2%醋酸鉛水溶液)和多肽組(添加2%多肽的基礎(chǔ)飼料,0.2%醋酸鉛水溶液),飼養(yǎng)60d后,觀察大鼠腦部海馬組織形態(tài)變化,測定海馬組織中鉛含量、camk2a、erk2和homer1mRNA的表達(dá)量。結(jié)果表明:鉛會(huì)導(dǎo)致海馬組織的細(xì)胞數(shù)量減少、細(xì)胞層變薄、細(xì)胞間隙增大、camk2a、erk2和homer1mRNA表達(dá)量極顯著降低;與染鉛模型組相比,多肽組海馬組織的鉛含量顯著降低、細(xì)胞數(shù)目增多、細(xì)胞核變大、細(xì)胞間隙縮小,同時(shí)海馬組織中NMDAR和mGluR5信號通路相關(guān)基因camk2a、erk2和homer1mRNA的表達(dá)量顯著或極顯著升高,表明刺芹側(cè)耳多肽可減輕鉛對大鼠海馬組織的損傷。
[Abstract]:In order to study the effect of Pleurotus eryngii polypeptide on brain tissue injury in lead exposed rats, male SD rats were randomly divided into normal control group (basal diet). Distilled water, lead exposed model group (basic diet 0.2% lead acetate aqueous solution) and polypeptide group (0.2% lead acetate solution supplemented with 2% polypeptide) were fed for 60 days, and the morphological changes of hippocampal tissue were observed. The content of lead in hippocampus and the expression of camk2a erk2 and homer1mRNA were measured. The results showed that lead could decrease the number of hippocampal cells, thin the cell layer, decrease the expression of camk2aerk2 and homer1mRNA, and decrease the content of lead and increase the number of cells in polypeptide group compared with the lead exposed model group. The expression of NMDAR and mGluR5 signaling pathway related genes camk2aerk2 and homer1mRNA were significantly or extremely increased in hippocampus, which indicated that polypeptide of Pleurotus prickly could attenuate the damage caused by lead in hippocampus of rats.
