巨癤蝙蛾不同蟲態(tài)的COI基因條形碼識別

【摘要】:【目的】巨癤蝙蛾Endoclita davidi(Poujade,1886)是雪峰蟲草菌Ophiocordyceps xuefengensis的重要寄主。本研究分析了該蟲線粒體DNA COI基因條形碼,以在不同蟲態(tài)及被蟲草菌寄生的僵蟲狀態(tài)下能快速準(zhǔn)確鑒定寄主種類!痉椒ā縋CR擴(kuò)增巨癤蝙蛾成蟲(足一對,胸肌,翅基部)、卵(單粒)、幼蟲(胸足一對)、蛹(少量體壁組織)及蟲草僵蟲(胸足或體壁)組織共計(jì)21個(gè)樣品的COI基因片段(約658 bp)并進(jìn)行測序和比對;以MEGA6.0軟件的K2-P雙參數(shù)模型計(jì)算種內(nèi)及種間遺傳距離;以鄰接法(neighbor-joining,NJ)和最大簡約法(maximum parsimony,MP)兩種方法構(gòu)建系統(tǒng)發(fā)育樹!窘Y(jié)果】聚類分析與形態(tài)學(xué)鑒定結(jié)果一致,所納入分析的20個(gè)樣品均為同一物種,節(jié)點(diǎn)支持率可達(dá)99%。種內(nèi)遺傳距離為0.00%~1.08%,平均0.54%。與蝙蝠蛾科其他6種昆蟲種間遺傳距離為5.36%~13.31%,平均10.47%;種間遺傳距離為種內(nèi)遺傳距離的19.39倍(10.47%vs 0.54%),而且種內(nèi)與種間遺傳距離沒有重疊區(qū)域!窘Y(jié)論】基于mtDNA COI基因DNA條形碼技術(shù)可以用于巨癤蝙蛾的快速準(zhǔn)確鑒定,并對蝙蝠蛾科(Hepialidae)昆蟲的分子系統(tǒng)發(fā)育分析有重要意義。
[Abstract]:[objective] Endoclita davidii Poujadea 1886 is an important host of Ophiocordyceps xuefengensis. In this study, the mitochondrial DNA COI gene bar code was analyzed, so that the host species could be identified quickly and accurately under different insect and parasitic conditions of Cordyceps. [methods] PCR was used to amplify adult adults (foot pair, pectoralis muscle) of Batacia megacephala (foot pair, pectoralis muscle). The COI gene fragments (about 658 BP) of 21 samples were obtained from the base of wings, eggs, larva (sternopoda pair), pupa (a small amount of body wall tissue) and Metarhizium anisopliae (chest foot or body wall). Phylogenetic tree was constructed by using K2-P two-parameter model of MEGA6.0 software and genetic distance between species and neighbor-joiningling NJs and maximum parsimony MP). [results] the results of cluster analysis and morphological identification were consistent. The 20 samples included in the analysis are all the same species, and the node support rate can reach 9910%. The intraspecific genetic distance was 0.001.08, with an average of 0.54. The genetic distance between species and other 6 species was 5.36, 13.31, with an average of 10.47, and the genetic distance between species was 19.39 times as high as that between species and 0.54.The genetic distance between species and species did not overlap. [conclusion] based on mtDNA COI gene DNA, the interspecific genetic distance was 19.39 times higher than that of other 6 species. [conclusion] there was no overlap between the interspecific genetic distance and the interspecific genetic distance. [conclusion] The form-coding technique can be used for rapid and accurate identification of Phasachis macrophylla. It is important to analyze the molecular phylogeny of Hepialidaeae.
