蘋果MdRGL3a基因的功能分析

【摘要】:為進一步揭示蘋果(Malus×domestica)中GA信號轉導機制以及MdDELLAs的生理功能,以‘嘎拉’蘋果為試材,克隆基因MdRGL3a(GenBank:DQ007887),并通過在擬南芥中過量表達,分析該基因的功能。qRT-PCR結果表明,MdRGL3a在蘋果不同組織中都有表達,其中在花和果實中表達量較高。在擬南芥中異位表達MdRGL3a,表現(xiàn)為根長和下胚軸變短,種子萌發(fā)較慢,葉片變小,植株矮化。過表達MdRGL3a擬南芥植株對赤霉素介導的生長反應敏感性降低,證明MdRGL3a是GA信號途徑中的一個負調控因子。
[Abstract]:In order to further reveal the mechanism of GA signal transduction and the physiological function of MdDELLAs in Malus 脳 domestica, the gene MdRGL3a was cloned into GenBank: DQ00788787, and was overexpressed in Arabidopsis thaliana. The results of qRT-PCR showed that MdRGL3a was expressed in different tissues of apple, especially in flower and fruit. The heterotopic expression of MdRGL3a in Arabidopsis thaliana showed that the root length and Hypocotyl became shorter, the seed germination was slower, the leaves were smaller and the plants were dwarfed. Overexpression of MdRGL3a in Arabidopsis thaliana plants is less sensitive to gibberellin mediated growth response, which suggests that MdRGL3a is a negative regulator in GA signaling pathway.
