蘋果MdRGL3a基因的功能分析

【摘要】:為進(jìn)一步揭示蘋果(Malus×domestica)中GA信號(hào)轉(zhuǎn)導(dǎo)機(jī)制以及MdDELLAs的生理功能,以‘嘎拉’蘋果為試材,克隆基因MdRGL3a(GenBank:DQ007887),并通過在擬南芥中過量表達(dá),分析該基因的功能。qRT-PCR結(jié)果表明,MdRGL3a在蘋果不同組織中都有表達(dá),其中在花和果實(shí)中表達(dá)量較高。在擬南芥中異位表達(dá)MdRGL3a,表現(xiàn)為根長和下胚軸變短,種子萌發(fā)較慢,葉片變小,植株矮化。過表達(dá)MdRGL3a擬南芥植株對赤霉素介導(dǎo)的生長反應(yīng)敏感性降低,證明MdRGL3a是GA信號(hào)途徑中的一個(gè)負(fù)調(diào)控因子。
[Abstract]:In order to further reveal the mechanism of GA signal transduction and the physiological function of MdDELLAs in Malus 脳 domestica, the gene MdRGL3a was cloned into GenBank: DQ00788787, and was overexpressed in Arabidopsis thaliana. The results of qRT-PCR showed that MdRGL3a was expressed in different tissues of apple, especially in flower and fruit. The heterotopic expression of MdRGL3a in Arabidopsis thaliana showed that the root length and Hypocotyl became shorter, the seed germination was slower, the leaves were smaller and the plants were dwarfed. Overexpression of MdRGL3a in Arabidopsis thaliana plants is less sensitive to gibberellin mediated growth response, which suggests that MdRGL3a is a negative regulator in GA signaling pathway.
