煙草中過表達(dá)丹參SAMDC基因增強(qiáng)其抗旱性(英文)

【摘要】:S-腺苷甲硫氨酸脫羧酶(SAMDC)是多胺合成代謝中的關(guān)鍵酶。本研究利用RT-PCR從丹參(Salvia miltiorrhiza)中獲得了全長(zhǎng)的SAMDC序列(Gen Bank No.GU983041.1),即SmSAMDC,與其他植物物種中的SAMDC氨基酸序列具有較高的同源性。為研究多胺在植物抗旱中的作用,利用根癌農(nóng)桿菌(Agrobacterium tumefaciens)介導(dǎo)的葉盤轉(zhuǎn)化法將SmSAMDC導(dǎo)入到煙草(Nicotiana tabacum)中,得到穩(wěn)定表達(dá)SmSAMDC的轉(zhuǎn)基因的T0和T1代煙草,用以抗干旱分析。SmSAMDC在煙草中的異源表達(dá)明顯地提高了轉(zhuǎn)基因T0和T1代煙草的抗旱能力。生理研究發(fā)現(xiàn),在干旱條件下,轉(zhuǎn)SmSAMDC煙草具有更高的相對(duì)含水量(relative water content,RWC)和抗氧化酶活性;相對(duì)較低的丙二醛(malondialdehyde,MDA)和超氧陰離子自由基(O2·-)含量。并且,在干旱脅迫環(huán)境中,SmSAMDC在煙草的過表達(dá)顯著地促進(jìn)了轉(zhuǎn)基因煙草中過氧化氫(H2O2)的積累。本研究證明過表達(dá)SmSAMDC能提高轉(zhuǎn)基因煙草的抗旱能力,為SmSAMDC在其他物種的應(yīng)用提供了理論依據(jù)。
[Abstract]:S-adenosine methionine decarboxylase (SAMDC) is a key enzyme in polyamine synthesis and metabolism. In this study, RT-PCR was used to obtain a full-length SAMDC sequence from Salvia miltiorrhizae, Gen. Bank No.GU983041. 1, or SmSAMDC, which has high homology with SAMDC amino acid sequences in other plant species. In order to study the effect of polyamines on drought resistance in plants, SmSAMDC was introduced into Nicotiana tabacum by Agrobacterium tumefaciens-mediated leaf disc transformation, and transgenic T _ 0 and T _ 1 tobacco with stable expression of SmSAMDC were obtained. The heterologous expression of .SmSAMDC in tobacco for drought resistance analysis significantly improved the drought resistance of transgenic T0 and T1 generation tobacco. Physiological studies showed that under drought conditions, transgenic SmSAMDC tobacco had higher relative water content (water) and antioxidant enzyme activity, lower malondialdehyde (malondialdehyde) and superoxide anion free radical (O 2 -) contents. Furthermore, the overexpression of SmSAMDC in transgenic tobacco significantly promoted the accumulation of H _ 2O _ 2 in transgenic tobacco under drought stress. This study proved that overexpression of SmSAMDC can improve the drought resistance of transgenic tobacco and provide a theoretical basis for the application of SmSAMDC in other species.
