‘翠冠’梨大果型芽變的細胞學(xué)及相關(guān)基因表達研究

【摘要】:【目的】對造成大果芽變的原因進行探討!痉椒ā恳浴涔凇捅憩F(xiàn)出大果性狀的芽變‘潘莊大翠冠’為材料,利用熒光AFLP分子標記進行基因組DNA序列差異分析;用流式細胞儀檢測‘潘莊大翠冠’的染色體倍性,用石蠟切片技術(shù)進行細胞學(xué)觀察,用實時熒光定量PCR檢測相關(guān)基因表達分析。【結(jié)果】AFLP分析證實‘潘莊大翠冠’為‘翠冠’的大果芽變,果實內(nèi)在品質(zhì)無明顯差異。流式細胞檢測結(jié)果顯示,‘潘莊大翠冠’為二倍體,與‘翠冠’相同!饲f大翠冠’梨的細胞分裂期從盛花期開始一直到花后24 d,較‘翠冠’長4 d,花后28 d時,果肉細胞層數(shù)顯著多于‘翠冠’。細胞周期蛋白D3(cyclin D3,CYCD3)在‘潘莊大翠冠’中的表達量顯著高于‘翠冠’。細胞周期蛋白A2(cyclin A2,CYCA2)、細胞周期蛋白依賴性激酶A1(cyclin-dependent kinase A1,CDKA1)和細胞周期蛋白依賴性激酶B2(cyclin-dependent kinase B2,CDKB2)等基因也有表達差異。【結(jié)論】‘潘莊大翠冠’確為‘翠冠’的大果芽變,其果實增大的機制并非染色體加倍,而是果實發(fā)育過程的細胞分裂期細胞的活躍增殖導(dǎo)致細胞分裂期的延長。
[Abstract]:[objective] to investigate the causes of granulation. [methods] Genomic DNA sequence difference was analyzed by fluorescence AFLP molecular marker using 'Cuiguan' and 'Panzhuang Dazuiguan' as materials. The chromosome ploidy of 'Panzhuang Daziguan' was detected by flow cytometry, and the cytology was observed by paraffin section technique. The expression of related genes was detected by real-time fluorescence quantitative PCR. [results] AFLP analysis confirmed that 'Panzhuang Dazuiguan' was the bud of 'Cuiguan', and there was no significant difference in the intrinsic quality of the fruit. The results of flow cytometry showed that 'Panzhuang Dazui Guan' was diploid, which was the same as' Cuiguan'. The cell division of 'Panzhuang Da Cuiguan' pear was 4 days longer than that of 'Cuiguan' from full flowering to 24 days after anthesis. At 28 days after anthesis, the number of cell layers in pulp was significantly more than that in 'Cuiguan'. The expression of cyclin D3(cyclin D _ 3 (CYCD _ 3) in 'Panzhuang D3' was significantly higher than that in 'Cuiguan'. There were also differences in the expression of cyclin A2(cyclin A2 (CYCA2), cyclin dependent kinase A1(cyclin-dependent kinase A1 (CDKA1) and cyclin dependent kinase B2(cyclin-dependent kinase B2 (CDKB2). [conclusion] 'Panzhuang Grand Cuiguan' is indeed the granulation of 'Cuiguan'. The mechanism of fruit enlargement is not chromosome doubling, but the prolongation of cell division due to the active proliferation of cells during fruit development.
