【摘要】：目的:探討白細(xì)胞介素-17(IL-17)血清表達(dá)水平及基因IL-17-197A/G位點(diǎn)單核苷酸多態(tài)性與乙型肝炎病毒(HBV)感染及臨床轉(zhuǎn)歸之間的關(guān)系。方法:以222例慢性HBV感染者作為病例組,以88例自限性HBV感染者為對(duì)照組。采用PCR結(jié)合熒光探針的體外DNA擴(kuò)增檢測(cè)技術(shù),定量檢測(cè)血清標(biāo)本中的HBV DNA含量;ELISA試劑盒檢測(cè)血清IL-17表達(dá)水平;采用貝克曼SNP分析儀,檢測(cè)IL-17-197A/G位點(diǎn)基因型及等位基因分布頻率。結(jié)果:各實(shí)驗(yàn)組IL-17表達(dá)水平差異具有統(tǒng)計(jì)學(xué)意義(F=158.1,r~2=0.61,P0.000 1)。L-17-197A/G位點(diǎn)AA、AG和GG基因型分布頻率在各組中的比較,差異具有統(tǒng)計(jì)學(xué)意義(χ~2=46.20,P0.000 1)。GG基因型在自限性感染組中分布頻率最高(78.41%),而在慢性乙肝肝硬化組中分布頻率最低(19.05%)。AA基因型在慢性乙型肝炎肝硬化組中分布頻率最高(67.72%)。結(jié)論:IL-17-197A/G位點(diǎn)AA基因型和A等位基因是河北漢族人群中HBV易感的宿主基因。
[Abstract]:Objective: to investigate the relationship between serum interleukin-17 (IL-17) expression level and single nucleotide polymorphism (SNP) of IL-17-197A/G site in patients with hepatitis B virus (HBV) infection and clinical outcome. Methods: 222 cases of chronic HBV infection were selected as case group and 88 cases of self limited HBV infection as control group. In vitro DNA amplification technique with PCR and fluorescence probe was used to quantitatively detect the HBV DNA content in serum samples and Elisa kit was used to detect the expression of serum IL-17, and Beckman SNP analyzer was used to detect the genotype and allelic frequency of IL-17-197A/G locus. Results: there were significant differences in the expression of IL-17 among the experimental groups. The distribution frequency of AA-AG and GG genotypes at the 1).L-17-197A/G locus AA-AG and GG in each group were statistically significant. The difference was statistically significant (蠂 ~ (2 +) ~ (46.20) P _ (0.000) 1).GG genotype) was the highest in the self-limited infection group (78.41%), but the lowest in the chronic hepatitis B cirrhosis group was 19.05%. AA genotype was the highest (67.72%) in the chronic hepatitis B cirrhosis group. Conclusion the AA genotype and A allele at the 1: 17-197 A / G locus are the host genes of susceptibility to HBV in Hebei Han population.
