水產(chǎn)品中6類抗生素抗性基因和Ⅰ類整合子的定量檢測

【摘要】:論文選取廣州市天河區(qū)兩家菜市場、一家海鮮市場和三家超市的48份水產(chǎn)品為研究對象,提取了三種不同水產(chǎn)品:蝦、魚和貝類的總DNA,采用實時熒光定量PCR(Quantitative Real-time PCR)法,對市售水產(chǎn)品中6類抗生素抗性基因(Antibiotic Resistance Genes,ARGs):tet A、tet B、tet M、sulⅡ、flo R、aph A-1、aad A、erm B、cml A和Ⅰ類整合子5’端整合酶基因int I1、3’端磺胺耐藥基因sulⅠ、季銨鹽化合物及溴化乙錠的耐藥基因qac EΔ1進行定量。研究顯示:6類ARGs及Ⅰ類整合子基因在水產(chǎn)品中均有檢出,基因拷貝數(shù)大小從10~2~10~8跨越七個數(shù)量級,最小值為1.86×10~2 copies/g,最大值為8.98×10~8 copies/g。三種水產(chǎn)品蝦、魚、貝類中,蝦類的ARGs含量最高,貝類最低;三類采樣地點中,菜市場來源的水產(chǎn)品中ARGs含量最高,海鮮市場來源的次之,超市來源的最低。本研究為進一步探索ARGs和可移動耐藥元件整合子介導(dǎo)的細菌耐藥性傳播而引起的食源性疾病機理奠定了基礎(chǔ)。
