百合雌蕊中雜交親和性與不親和性差異表達(dá)基因分析

【摘要】:東方百合與卷瓣組野生百合雜交(簡(jiǎn)稱OS組合),屬于親緣關(guān)系最遠(yuǎn)的組間雜交,雜交障礙最大。挖掘雌蕊中雜交親和性與不親和性差異表達(dá)基因,有可能解釋不親和性雜交中引起花粉管定向生長(zhǎng)異常的機(jī)理,進(jìn)而闡明不親和性雜交的分子機(jī)理,為實(shí)施克服雜交不親和障礙技術(shù)措施提供依據(jù)。本研究采用抑制消減雜交技術(shù)(SSH技術(shù)),分別以不親和性雜交和親和性雜交的東方百合雌蕊為試驗(yàn)組(tester)和驅(qū)動(dòng)組(driver),建立正向抑制消減雜交文庫(kù),隨機(jī)挑選180個(gè)陽(yáng)性克隆,經(jīng)過(guò)測(cè)序、去劣、去冗余、序列比對(duì),有113個(gè)EST與數(shù)據(jù)庫(kù)中的已知序列具有同源性,最終獲得10個(gè)差異表達(dá)基因。采用RT-PCR技術(shù)對(duì)10個(gè)差異表達(dá)基因進(jìn)行驗(yàn)證,其中有1個(gè)基因在不親和性雜交的雌蕊中表達(dá)上調(diào)、7個(gè)基因表達(dá)下調(diào)、2個(gè)基因無(wú)明顯變化。對(duì)差異表達(dá)基因進(jìn)行功能注釋及分類,差異表達(dá)基因主要集中于信號(hào)轉(zhuǎn)導(dǎo)(包括絲氨酸/蘇氨酸蛋白磷酸酶PP2A-3、鈣依賴蛋白激酶CDPK、小G結(jié)合蛋白)、抗逆防御(包括分子伴侶(clp B)、過(guò)氧化氫酶CAT2)、轉(zhuǎn)運(yùn)(包括質(zhì)膜ATPase、焦磷酸酶質(zhì)子泵)、蛋白命運(yùn)(包括60S核糖體蛋白)等功能類別。據(jù)此推測(cè),在東方百合×山丹組(系)間不親合性雜交育種過(guò)程中,可能引起雌蕊與花粉管信號(hào)交流異常、物質(zhì)轉(zhuǎn)運(yùn)能力減弱和雌蕊耐受逆境能力降低,從而導(dǎo)致雜交不親和性。
[Abstract]:Hybrid between Oriental Lilium and Wild Lilium curl group (abbreviated as OS combination) is the most distant cross, and the cross obstacle is the biggest.Digging out the differentially expressed genes between incompatibility and incompatibility in pistil may explain the mechanism of pollen tube directional growth abnormality in incompatibility hybridization, and then elucidate the molecular mechanism of incompatibility hybridization.To provide the basis for the implementation of technical measures to overcome the barrier of cross incompatibility.In this study, the suppression subtractive hybridization (SSH) technique was used to establish a positive suppression subtractive hybridization library, and 180 positive clones were randomly selected.After sequencing, inferiority, redundancy removal, sequence alignment, 113 EST have homology with known sequences in the database, and finally 10 differentially expressed genes were obtained.Ten differentially expressed genes were verified by RT-PCR technique. Among them, 1 gene was up-regulated in pistil of incompatibility hybridization, 7 genes were down-regulated, and 2 genes had no obvious change.Functional annotation and classification of differentially expressed genes,Differentially expressed genes are mainly concentrated in signal transduction (including serine / threonine protein phosphatase PP2A-3, calcium-dependent protein kinase (CDPKK), small G-binding protein, anti-stress defense (including molecular chaperone, catalase CAT2, and transport).Plasma membrane Pase, pyrophosphatase proton pump, protein fate (including 60s ribosomal protein) and other functional categories.It is inferred that, in the process of incompatibility hybrids between Lilium oriental 脳 Shandan group (lines), it may lead to abnormal signal exchange between pistil and pollen tube, decrease of material transport ability and decrease of pistil tolerance to stress.This leads to cross incompatibility.
