飼料浮萍水平對(duì)大正三色錦鯉TYR及MC1R基因表達(dá)的影響

【摘要】:為探究飼料中不同浮萍添加水平對(duì)大正三色錦鯉酪氨酸酶TYR以及黑素皮質(zhì)素受體MC1R基因表達(dá)的影響,配制浮萍添加水平分別為0%、3%、6%、9%、13%、14%6種等蛋白(38.6%)飼料飼養(yǎng)大正三色錦鯉(42.19±1.32)g 8周,分別記為Diet 1(0/13)、Diet 2(3/10)、Diet 3(6/7)、Diet 4(9/4)、Diet 5(13/1)、Diet 6(14/0)。每種飼料設(shè)3個(gè)重復(fù),每個(gè)重復(fù)飼喂10尾魚。試驗(yàn)結(jié)果表明:(1)TYR基因在大正三色錦鯉的心臟、肝臟、眼睛、黑色皮膚、腦中均有表達(dá),并且在腦、皮膚、眼睛中表達(dá)量相對(duì)較高;(2)各組織中TYR mRNA表達(dá)量隨浮萍添加水平的升高均有一定波動(dòng),在皮膚與眼睛中,以Diet 6(14/0)組顯著高于其他各組(P0.05),在腦中,以Diet 3(6/7)組顯著高于除Diet 6的其他各組(P0.05);(3)皮膚與眼睛中MC1R mRNA表達(dá)量以Diet 6(14/0)組顯著高于其他各組(P0.05),而在腦中各組差異均不顯著(P0.05)。綜上所述,在本試驗(yàn)條件下,飼料不同浮萍添加水平促進(jìn)大正三色錦鯉TYR以及MC1R基因表達(dá),有利于魚體黑色素沉積,因此選擇Diet 6(14/0)組飼料投喂大正三色錦鯉最適。
[Abstract]:In order to investigate the effect of different levels of duckweed supplementation on the expression of tyrosinase TYR and melanin receptor MC1R gene in tricolor koi carp, the supplementation level of duckweed was 0 + 3X and 131313146, respectively. The feeding level was 42.19 鹵1.32g / g for 8 weeks.It's called Diet 1 / 13 / 13 / 2 / 2 / 10 / 3 / 7 / 7 / 4 / 4 / 4 / 5 / 5 / 6 / 614 / 0 / 0 respectively.Each feed was fed with 3 repeats and 10 fish were fed with each repeats.The results showed that the TYR gene was expressed in the heart, liver, eyes, black skin and brain of the tricolor carp, and was also expressed in the brain and the skin.The expression of TYR mRNA fluctuated with the increase of duckweed supplementation level. In the skin and eyes, the expression of TYR mRNA in the skin and eyes was significantly higher than that in the other groups (P 0.05), and in the brain, the expression of TYR mRNA in the skin and eyes was significantly higher than that in the other groups.The expression of MC1R mRNA in the skin and eyes was significantly higher in the Diet 3 / 6 / 7 group than in the other groups except Diet 6. The expression of MC1R mRNA in the skin and eyes was significantly higher in the Diet 614 / 0 group than in the other groups, but no significant difference was found in the brain.To sum up, under the experimental conditions, different feeding levels of duckweed promoted the expression of TYR and MC1R genes in tricolor koi, which was beneficial to the melanin deposition in fish body. Therefore, it was most suitable to choose the feed of Diet 6 / 14 / 0) group to feed Dazheng tricolor koi.
