茶樹(shù)發(fā)育相關(guān)基因CsARGOS的克隆與表達(dá)特性分析

【摘要】:植物發(fā)育相關(guān)基因ARGOS基因受生長(zhǎng)素誘導(dǎo),在植物的生長(zhǎng)發(fā)育和激素調(diào)控方面起著重要的作用。本實(shí)驗(yàn)選取茶樹(shù)品種‘龍井43’作為試驗(yàn)材料,利用RT-PCR方法,從茶樹(shù)c DNA中克隆得到茶樹(shù)ARGOS基因,命名為Cs ARGOS。序列分析表明,Cs ARGOS基因開(kāi)放閱讀框?yàn)?66 bp,編碼121個(gè)氨基酸。進(jìn)化樹(shù)分析顯示,Cs ARGOS蛋白與雙子葉植物的進(jìn)化樹(shù)關(guān)系相對(duì)較近,與單子葉植物的進(jìn)化樹(shù)關(guān)系相對(duì)較遠(yuǎn)。序列多重比對(duì)顯示,Cs ARGOS蛋白具有ARGOS蛋白的特征基序,C端均含有豐富的亮氨酸,N端親水性氨基酸居多,這些區(qū)域在物種中是相對(duì)保守的。氨基酸組分、理化性質(zhì)、親水性/疏水性、二級(jí)結(jié)構(gòu)預(yù)測(cè)和無(wú)序化分析顯示,茶樹(shù)Cs ARGOS是疏水性蛋白,以隨機(jī)卷曲為主,無(wú)序化特征不明顯。利用實(shí)時(shí)熒光定量PCR對(duì)Cs ARGOS基因在茶樹(shù)中的表達(dá)進(jìn)行檢測(cè),分析結(jié)果表明在不同組織中,Cs ARGOS基因的表達(dá)水平從高到低依次為根第三葉第四葉老葉嫩莖第二葉第一葉;不同激素處理下,Cs ARGOS的表達(dá)水平從高到低依次為GA_3SAABAMe JAIAA。
[Abstract]:Plant development related gene ARGOS gene is induced by auxin and plays an important role in plant growth and hormone regulation.In this experiment, we selected the tea variety 'Longjing 43' as the experimental material, and cloned the ARGOS gene of tea tree from c DNA by RT-PCR method, named as Cs ARGOSs.Sequence analysis showed that the open reading frame of Cs ARGOS gene was 366 BP, encoding 121 amino acids.The phylogenetic tree analysis showed that the relationship between Cs ARGOS protein and the evolutionary tree of dicotyledonous plants was relatively close to that of monocotyledonous plants.Sequence multiple alignment showed that the C terminal of ARGOS protein had the characteristic of ARGOS protein, most of which were rich in leucine N-terminal hydrophilic amino acids, and these regions were relatively conserved in species.Amino acid composition, physicochemical properties, hydrophilicity / hydrophobicity, secondary structure prediction and disordered analysis showed that Cs ARGOS was hydrophobic protein, mainly in random curl.The expression of Cs ARGOS gene in tea plants was detected by real-time fluorescence quantitative PCR. The results showed that the expression level of Cs ARGOS gene in different tissues was the first leaf of the second leaf of the tender stem of the fourth leaf of the third leaf of root and the first leaf of the second leaf of the tender stem of the fourth leaf of the root in the order of high to low.The expression level of Cs ARGOS in different hormone treatments was GA_3SAABAMe JAIAA from high to low.
