【摘要】：目的對(duì)ABO正反定型不符的幼兒標(biāo)本進(jìn)行血清學(xué)鑒定和基因檢測(cè)分析。方法常規(guī)血清學(xué)檢測(cè),PCR-SSP進(jìn)行ABO基因初步分型,再使用ABO血型-A亞型基因分型試劑盒進(jìn)行A亞型分型鑒定。結(jié)果血清學(xué)結(jié)果顯示標(biāo)本正定型為A型(抗A:1+~W),反定為O型(Ac2+,Bc4+);PCR-SSP A亞型分型鑒定結(jié)果顯示標(biāo)本基因型為Ax14/O2型。結(jié)論疑難血型鑒定有時(shí)需要結(jié)合血清學(xué)和分子生物學(xué)方法才能準(zhǔn)確定型,此例血清學(xué)正反定型不符標(biāo)本表型為Ax,其基因型為Ax14/O2。
[Abstract]:Objective to study the serological identification and gene analysis of ABO samples.Methods ABO genotyping was carried out by PCR-SSP, and A subtype was identified by using ABO blood group A subtype genotyping kit.Results the serological results showed that the positive type of the specimen was A type (anti-A: 1) and the positive type was O type Ac2 Bc4 and PCR-SSP A subtype. The results showed that the genotype of the specimen was Ax14/O2 type.Conclusion the identification of difficult blood groups sometimes requires the combination of serological and molecular biological methods in order to identify accurately. This case of serological positive and negative typing does not match the phenotype of Ax14 / O2, and its genotype is Ax14 / O2.
