【摘要】：該文旨在探討沉默信息調(diào)節(jié)因子6(silent information regulator 6,SIRT6)基因沉默對裸鼠異位移植人肝癌細(xì)胞增殖和凋亡的影響。構(gòu)建穩(wěn)定細(xì)胞系SIRT6-sh RNA-SK-Hep-1和sh Cont-SKHep-1,并應(yīng)用定量逆轉(zhuǎn)錄PCR(q RT-PCR)和Western blot檢測SIRT6基因的表達(dá)水平;將穩(wěn)定轉(zhuǎn)染細(xì)胞注入裸鼠皮下,實時監(jiān)測裸鼠移植瘤的生長,7周后剝離出裸鼠移植瘤并稱重;免疫組織化學(xué)分析SIRT6、Ki-67的蛋白質(zhì)水平。進(jìn)一步應(yīng)用q RT-PCR檢測SIRT6基因沉默對下游靶向分子凋亡抑制蛋白(inhibitor of apoptosis proteins,IAPs)家族的影響;Western blot檢測X連鎖凋亡抑制蛋白(X-linked inhibitor of apoptosis protein gene,XIAP)和多腺苷二磷酸核糖聚合酶(poly ADP-ribose polymerase,PARP)蛋白質(zhì)水平。結(jié)果顯示,成功構(gòu)建了沉默SIRT6基因的SK-Hep-1穩(wěn)定細(xì)胞系;SIRT6-sh RNASK-Hep-1組裸鼠移植瘤體積和質(zhì)量較sh Cont-SK-Hep-1組均減少(P0.01);免疫組織化學(xué)發(fā)現(xiàn),SIRT6-sh RNA-SK-Hep-1組Ki-67水平下調(diào);沉默SIRT6基因下調(diào)XIAP m RNA和蛋白質(zhì)水平,并增加PARP蛋白質(zhì)的剪切水平。該研究結(jié)果提示,SIRT6基因沉默可能通過下調(diào)XIAP的表達(dá)抑制裸鼠異位移植人肝癌細(xì)胞的生長。
[Abstract]:The aim of this study was to investigate the effect of silencing of 6(silent information regulator 6 sir6) gene silencing on proliferation and apoptosis of heterotopic transplanted human hepatoma cells in nude mice. Stable cell lines SIRT6-sh RNA-SK-Hep-1 and sh Cont-SKHep-1 were constructed and detected by quantitative reverse transcription PCR(q RT-PCR and Western blot. The expression level of SIRT6 gene; The stable transfected cells were injected subcutaneously into nude mice, and the growth of xenografts in nude mice was monitored in real time. After 7 weeks, the xenografts were stripped out and weighed. The protein level of SIRT6KI-67 was analyzed by immunohistochemistry. The effect of SIRT6 gene silencing on the downstream inhibitor of apoptosis proteinsi IAPs was further detected by Q RT-PCR. Western blot was used to detect X-linked inhibitor of apoptosis protein. Protein levels of gene XIP and poly ADP-ribose polymerase of polyadenosine diphosphate. SK-Hep-1 stable cell line with silencing SIRT6 gene was successfully constructed. The volume and mass of xenografts in the RNASK-Hep-1 group were lower than those in the sh Cont-SK-Hep-1 group, and the Ki-67 level was down-regulated in the SIRT6-sh RNA-SK-Hep-1 group, and the XIAP m RNA and protein levels were down-regulated by silencing SIRT6 gene. The results suggested that the silencing of SIRT6 gene might inhibit the growth of xenotransplantation of human hepatoma cells by down-regulating the expression of XIAP in nude mice.
