絲切蛋白CFL2基因低表達(dá)對(duì)小鼠成肌細(xì)胞C2C12肌纖維信號(hào)通路關(guān)鍵成分的影響

【摘要】:探討低表達(dá)絲切蛋白CFL2基因?qū)π∈蟪杉〖?xì)胞C2C12信號(hào)通路關(guān)鍵成分的影響。采用RNAi技術(shù),將豬CFL2基因真核表達(dá)shRNA重組體穩(wěn)定轉(zhuǎn)染至小鼠成肌細(xì)胞C2C12后提取其總RNA和總蛋白質(zhì),采用Real-time PCR和Western blot檢測(cè)CFL2基因低表達(dá)后成肌細(xì)胞C2C12信號(hào)通路關(guān)鍵成分(MEF2C、sk MLCK、Rho A、AMPKα2、p38和Ca M)的表達(dá)情況。結(jié)果表明:在3個(gè)穩(wěn)轉(zhuǎn)細(xì)胞株中MEF2C、Ca M、p38和AMPKα2的mRNA表達(dá)均為極顯著下調(diào)(P0.01),sk MLCK的mRNA表達(dá)為極顯著上調(diào)(P0.01),其它成分不顯著;MEF2C和Ca M的蛋白表達(dá)在3個(gè)穩(wěn)轉(zhuǎn)細(xì)胞株中均為顯著或極顯著下調(diào)(P0.05或P0.01),其他4種成分(p38、Rho A、AMPKα2和sk MLCK)均差異不顯著(P0.05),說明CFL2低表達(dá)顯著下調(diào)信號(hào)通路關(guān)鍵成分MEF2C和Ca M的表達(dá),CFL2與Ca M和sk MLCK分別呈顯著正相關(guān)和負(fù)相關(guān)。
[Abstract]:To investigate the effect of low expression of CFL2 gene on the key components of C2C12 signaling pathway in mouse myoblasts, the porcine CFL2 gene eukaryotic expression shRNA recombinant was stably transfected into mouse myoblast C2C12 to extract its total RNA and total protein by RNAi technique. Real-time PCR and Western blot were used to detect the expression of the key components of C2C12 signal pathway in myoblasts after low expression of CFL2 gene. The results showed that the mRNA expression of MEF2CU Ca Mp38 and AMPK 偽 2 were significantly down-regulated in three stable cell lines. The mRNA expression of P0.01 MLCK was significantly up-regulated, while the protein expression of MEF2C and Cam was significantly or extremely down-regulated in all of the three stable cell lines, while the other four components, p38, Rho, AMPK 偽 2, and SK MLCK, had no significant difference in the expression of P0.05, said P0.05, saying that the expression of MEF2C and Cam in the three stable cell lines was significantly or extremely down-regulated by P0.01 or P0.01a, and that there was no significant difference between the other four components, P38, Rho, AMPK 偽 2 and SK MLCKs, said. The low expression of CFL2 down-regulated the expression of MEF2C and Cam. The expression of CFL2 was positively and negatively correlated with Cam and SK MLCK, respectively.
