綿羊MUSTN1基因的克

【摘要】:為了探討綿羊(Ovis aires)MUSTN1基因特征及其在不同發(fā)育階段的背最長肌與股二頭肌組織中的表達規(guī)律,選取0、2、6、12和24月齡健康綿羊15只,每組3只,采取背最長肌和股二頭肌組織,應(yīng)用分子生物學(xué)等方法,對MUSTN1基因CDS區(qū)進行克隆,對其進行生物信息學(xué)和組織表達分析。結(jié)果表明,MUSTN1基因CDS區(qū)全序列為248 bp,編碼82個氨基酸,其分子量為8.89 ku,等電點為9.93,GC含量大于AT含量,含有5個磷酸化位點、5個N-糖基化位點、1個O-糖基化位點、0個跨膜結(jié)構(gòu)、疏水性蛋白,二級結(jié)構(gòu)主要以無規(guī)卷曲為主;與山羊(Capra hircus)該基因核苷酸以及編碼氨基酸序列的同源性最高,分別為99.2%、100%,其次為牛(Bos taurus),分別為97.2%、100%。MUSTN1基因在綿羊不同發(fā)育階段(0、2、6、12和24月齡)的背最長肌與股二頭肌中均有表達,在背最長肌中,24月齡MUSTN1表達量最高,極顯著高于其他月齡(P0.01),0月齡MUSTN1表達量最低,極顯著低于2、12月齡(P0.01),顯著低于6月齡(P0.05);而在6月齡綿羊股二頭肌中MUSTN1表達量最高且極顯著高于其他月齡(P0.01),24月齡表達量次之且極顯著高于12月齡(P0.01),顯著高于0月齡(P0.05)。結(jié)論為綿羊MUSTN1基因的編碼區(qū)序列在物種間具有保守性,且在不同發(fā)育階段的背最長肌和股二頭肌組織中均表達,其表達量的高低可能與骨骼肌肉的生長快慢相關(guān)。
[Abstract]:In order to investigate the characteristics of Ovis aires)MUSTN1 gene and its expression in the longissimus dorsi and biceps femoris at different developmental stages, 15 healthy sheep of 12 and 24 months of age were selected, 3 in each group, and 3 in each group. The longissimus dorsi muscle and biceps femoris muscle were used. The CDS region of MUSTN1 gene was cloned by molecular biology, and its bioinformatics and tissue expression analysis were carried out. The results showed that the whole sequence of CDS region of MUSTN1 gene was 248bp, encoding 82 amino acids. Its molecular weight is 8.89 ku. the content of isoelectric point is 9.93% GC is higher than that of AT, and it contains 5 phosphorylation sites, 5 N-glycosylation sites, 1 O-glycosylation site, 0 transmembrane structure, hydrophobic protein and secondary structure. The nucleotide sequence and amino acid sequence of Capra hircus gene had the highest homology, 99.2 and 100, respectively, followed by Bos taurusus, which was 97.2and 100.MUSTN1 gene was expressed in the longissimus dorsi muscle and biceps femoris muscle at different developmental stages of 12 and 24 months of age, respectively. In the longissimus dorsi muscle, the highest expression of MUSTN1 was observed at 24 months old, which was significantly higher than that in the other months of age. The expression of MUSTN1 in the biceps femoris femoris was the highest and significantly higher in the biceps femoris than that in the other months of age, and was significantly higher than that of the 12-month-old P0.01, and was significantly higher than that of the 12-month-old P0.01M, and was significantly higher than that of the 0-month-old sheep, and was significantly lower than that of the 6-month-old sheep's biceps femoris (P0.01), which was significantly higher than that of the 6-month-old sheep's biceps femoris. Conclusion the coding region of sheep MUSTN1 gene is conserved among species. The expression level of longissimus dorsi muscle and biceps femoris muscle may be related to the growth rate of skeletal muscle.
