中間錦雞兒CiPP2C8-like基因的克隆及表達(dá)分析

【摘要】:蛋白磷酸酶PP2C作為調(diào)節(jié)因子直接或間接調(diào)控逆境脅迫信號(hào)途徑和生長(zhǎng)發(fā)育過(guò)程。本研究從已構(gòu)建好的抑制性消減文庫(kù)(SSH)中獲得1條PP2C基因序列,對(duì)其進(jìn)行了克隆。測(cè)序表明該基因c DNA長(zhǎng)為999 bp,共編碼333個(gè)氨基酸,推導(dǎo)該蛋白分子量為36.12 k D,等電點(diǎn)為6.71,是一種親水蛋白。經(jīng)氨基酸序列比對(duì)和進(jìn)化樹(shù)分析表明,該基因?qū)儆赑P2C家族成員,并命名為Ci PP2C8-like。實(shí)時(shí)熒光定量PCR檢測(cè)表明中間錦雞兒Ci PP2C8-like基因表達(dá)受鹽和脫水脅迫誘導(dǎo)。
[Abstract]:As a regulatory factor, protein phosphatase (PP2C) directly or indirectly regulates stress signaling pathway and growth and development. In this study, a PP2C gene sequence was obtained from the constructed suppression subtractive library. The cDNA was cloned. The result of sequencing showed that the gene was 999bplong, encoding 333amino acids, deduced that the molecular weight of the protein was 36.12kD and the isoelectric point was 6.71.The results of amino acid sequence alignment and phylogenetic tree analysis showed that the protein was hydrophilic. The gene belongs to the PP2C family and is named Ci PP2C8-like. The real-time fluorescence quantitative PCR analysis showed that the expression of ci PP2C8-like gene in Caragana intermedia was induced by salt and dehydration stress.
