中國甘薯雙生病毒C4基因分子變異及遺傳多態(tài)性分析

【摘要】:甘薯雙生病毒是中國甘薯上一種重要的病毒,其C4基因編碼的蛋白是一個(gè)多功能蛋白,參與了寄主癥狀的形成及病毒在植物體內(nèi)移動(dòng)等。本研究利用PCR方法擴(kuò)增了24個(gè)分離物的C4基因,通過SDT、Dna SP等軟件分析,結(jié)果表明,24個(gè)分離物核苷酸序列均為258個(gè)堿基,一致率為73.3%~100.0%,編碼85個(gè)氨基酸,一致率為47.1%~100.0%。中國甘薯雙生病毒分離物C4基因構(gòu)建系統(tǒng)進(jìn)化樹分為2個(gè)組,組Ⅰ包括本研究的15個(gè)雙生病毒分離物及中國大部分雙生病毒分離物,又分為2個(gè)亞組,亞組2僅有15個(gè)雙生病毒分離物,其中本研究的6個(gè)分離物位于此亞組;組Ⅱ含有30個(gè)雙生病毒分離物,包括本研究的9個(gè)分離物。非同義突變與同義突變比值大于1,說明C4基因處于正向選擇。亞組1錯(cuò)配分布圖為單峰,說明該組種群呈突然暴發(fā)的狀態(tài),其余組或亞組為多峰鋸齒狀,表明種群或者亞種群已經(jīng)存在很長時(shí)間,長期處于動(dòng)態(tài)平衡。
[Abstract]:Sweet potato twin virus is an important virus in Chinese sweet potato. The protein encoded by C4 gene is a multifunctional protein. The PCR method was used to amplify the C4 gene of 24 isolates. The results showed that the nucleotide sequences of 24 isolates were 258 bases. The concordance rate was 73.3%, encoding 85 amino acids, and the concordance rate was 47.1% and 100.0.The phylogenetic tree of Chinese sweet potato Gemini virus isolate C4 gene was divided into two groups. Group I included 15 isolates of Geminiviruses and most of the isolates of Chinese Gemini virus. There were only 15 isolates from subgroup 2, 6 of which were located in this subgroup, and group 鈪,
