外源水楊酸誘導(dǎo)的部分甜瓜自毒作用相關(guān)基因的表達(dá)分析

【摘要】:以甜瓜(Cucumis melo)品種‘新銀輝’為材料,采取根灌法,以水楊酸(10μmol/L)處理甜瓜幼苗,通過(guò)Real-Time PCR對(duì)葉片中部分自毒作用相關(guān)基因的表達(dá)量進(jìn)行定量分析,以探究外源水楊酸誘導(dǎo)對(duì)甜瓜自毒作用的影響。結(jié)果顯示,經(jīng)外源水楊酸誘導(dǎo)后:(1)在催化苯丙烷類(lèi)代謝途徑的3個(gè)初始反應(yīng)的蛋白酶編碼基因中,PAL和C4H隨處理后時(shí)間增加,呈逐漸上調(diào)趨勢(shì),但PAL比C4H更快響應(yīng)脅迫,而4CL轉(zhuǎn)錄水平?jīng)]有顯著變化;(2)在類(lèi)黃酮生物合成的3個(gè)關(guān)鍵酶的基因中,CHI、F3H均顯示出不同程度的下調(diào),CHS于處理后1 d略微上調(diào),然后恢復(fù)至處理前水平;(3)兩個(gè)轉(zhuǎn)錄因子基因WD40、R2R3-MYB分別在不同處理時(shí)間后顯著下調(diào);(4)WD40與CHI的表達(dá)相關(guān)性較高。據(jù)上述結(jié)果 ,水楊酸誘導(dǎo)后:(Ⅰ)CHI表達(dá)可能與轉(zhuǎn)錄因子WD40蛋白相關(guān);(Ⅱ)WD40、R2R3-MYB與PAL、C4H、4CL、CHS間不存在簡(jiǎn)單、明確的調(diào)控關(guān)系;(Ⅲ)柚皮苷、表兒茶素、蘆丁等甜瓜主要化感物質(zhì)合成減少。
[Abstract]:The seedling of muskmelon was treated with 10 渭 mol / L salicylate by root irrigation with Cucumis meloides as material. The expression of some autotoxic genes in leaves was quantitatively analyzed by Real-Time PCR. The effects of exogenous salicylic acid on autotoxicity of muskmelon were studied. The results showed that the effects of exogenous salicylic acid on the autotoxicity of muskmelon were investigated. The results showed that the expression of pal and C4H in the three protease coding genes that catalyze the metabolic pathway of phenylpropanes increased with the treatment of exogenous salicylic acid. However, the transcription level of 4CL did not change significantly. The genes of three key enzymes of flavonoid biosynthesis showed different down-regulation of CHII-F3H, which was slightly up-regulated on the 1st day after treatment, but PAL was more responsive to stress than that of C4H, but the transcription level of 4CL was not changed significantly. Two transcription factor genes, WD40, R2R3-MYB, significantly down-regulated the expression of WD40, R2R3-MYB and CHI, respectively.
