ApoE基因敲除小鼠海馬CA3區(qū)神經(jīng)元形態(tài)學(xué)和微管相關(guān)蛋白2表達(dá)變化

【摘要】:目的觀察載脂蛋白E基因敲除(Apo E KO)對(duì)小鼠海馬CA3區(qū)神經(jīng)元形態(tài)結(jié)構(gòu)改變以及微管相關(guān)蛋白2(MAP-2)表達(dá)的影響。方法 10只野生型C57BL/6J小鼠為對(duì)照組,10只Apo E基因敲除小鼠為KO組,普通飼料喂養(yǎng)3個(gè)月。采用分光光度計(jì)檢測(cè)血脂變化;取兩組小鼠腦組織,HE染色和電鏡技術(shù)觀察海馬CA3區(qū)結(jié)構(gòu)改變,免疫組織化學(xué)技術(shù)觀察兩組小鼠海馬CA3區(qū)MAP-2表達(dá)變化,采用計(jì)算機(jī)圖像分析系統(tǒng)檢測(cè)其平均光密度(MOD)值。結(jié)果與對(duì)照組比較,KO組小鼠血漿總膽固醇、血漿三酰甘油、低密度脂蛋白含量明顯升高(P0.05,P0.01)。光鏡下,KO組小鼠海馬CA3區(qū)錐體細(xì)胞較小,排列稀疏松散。電鏡下,KO組海馬神經(jīng)元內(nèi)線粒體腫脹變性,嵴消失;神經(jīng)纖維髓鞘松懈;突觸小泡數(shù)量減少,突觸間隙模糊等。免疫組織化學(xué)結(jié)果顯示,對(duì)照組小鼠海馬神經(jīng)元內(nèi)MAP-2高表達(dá),KO組含量明顯減少;圖像分析顯示KO組CA3區(qū)MAP-2MOD值明顯降低(P0.05)。結(jié)論 Apo E KO小鼠海馬神經(jīng)元結(jié)構(gòu)出現(xiàn)病理改變,MAP-2表達(dá)下降,提示Apo E KO與神經(jīng)退行性病變有一定聯(lián)系。
[Abstract]:Objective to observe the effects of apolipoprotein E knockout (Apo E KOO) on the changes of neuronal morphology and the expression of microtubule-associated protein 2mMAP-2.Methods Ten wild-type C57BL / 6J mice were used as control group and 10 Apo E knockout mice were used as KO group. The changes of serum lipids were detected by spectrophotometer, the changes of CA3 structure in hippocampus were observed by HE staining and electron microscopy, and the changes of MAP-2 expression in hippocampal CA3 were observed by immunohistochemical technique. The average optical density (MOD) was measured by computer image analysis system. Results compared with the control group, the plasma total cholesterol and triglyceride were measured in the KO group. The content of low density lipoprotein (LDL) increased significantly in P0.05P0.010.The pyramidal cells in the hippocampal CA3 region of KO group were smaller and loosely arranged. Under electron microscope, mitochondria swelled and degenerated in hippocampal neurons, and the ridges disappeared, and the myelin sheath of nerve fibers was lax. The number of synaptic vesicles was decreased and the synaptic gap was blurred. The results of immunohistochemistry showed that the content of MAP-2 in hippocampal neurons of control group was significantly decreased. Image analysis showed that the MAP-2MOD value of CA3 in KO group was significantly lower than that of P0.05.Conclusion the expression of MAP-2 in hippocampal neurons of Apo E KO mice is decreased, suggesting that Apo EKO is associated with neurodegenerative lesions.
