孕前應(yīng)激誘導(dǎo)的產(chǎn)后抑郁模型小鼠海馬精神分裂癥斷裂基因1的表達(dá)變化

【摘要】:目的以孕前應(yīng)激誘導(dǎo)方式建立產(chǎn)后抑郁癥(PPD)小鼠模型,評(píng)估母鼠產(chǎn)后的異常行為,檢測(cè)母鼠海馬精神分裂癥斷裂基因1(DISC1)及血清雌二醇、皮質(zhì)酮的變化,揭示產(chǎn)后抑郁的發(fā)病機(jī)制。方法將32只Balb/c母鼠采用隨機(jī)數(shù)字表法分為對(duì)照組和模型組,每組16只,對(duì)照組小鼠不給予任何刺激,模型組小鼠給予慢性束縛刺激。3周后對(duì)照組和模型組均予雌雄合籠交配懷孕,分娩后即為對(duì)照組和孕前應(yīng)激誘導(dǎo)PPD模型組。于分娩后3周對(duì)母鼠進(jìn)行曠場(chǎng)測(cè)試、懸尾測(cè)試及糖水偏愛(ài)測(cè)試以評(píng)價(jià)造模是否成功。取海馬檢測(cè)DISC1基因及蛋白的表達(dá),眼眶取血檢測(cè)血清雌二醇及皮質(zhì)酮水平。結(jié)果產(chǎn)后3周,與對(duì)照組相比,PPD模型組在曠場(chǎng)測(cè)試中自發(fā)活動(dòng)無(wú)改變(P0.05),在懸尾測(cè)試中的絕望不動(dòng)時(shí)間顯著增加(t=-4.950,P0.001),糖水偏愛(ài)顯著降低(t=2.475,P0.05)。與對(duì)照組相比,PPD模型組海馬DISC1基因(t=-8.915,P0.001)及蛋白(t=-5.004,P0.01)表達(dá)均顯著上調(diào),而血清皮質(zhì)酮及雌二醇均無(wú)變化(P均0.05)。結(jié)論慢性孕前應(yīng)激可誘導(dǎo)Balb/c母鼠表現(xiàn)出產(chǎn)后抑郁樣癥狀,其發(fā)病機(jī)制可能與調(diào)控海馬DISC1的基因和蛋白表達(dá)有關(guān)。
[Abstract]:Objective to establish the model of postpartum depression (PPD) mice by pre-pregnancy stress, to evaluate the abnormal behavior of postpartum rats, and to detect the changes of estradiol and corticosterone in the serum of the female rats. Methods 32 Balb/c mice were randomly divided into control group (n = 16) and model group (n = 16). The model group mice were given chronic restraint stimulation for 3 weeks, the control group and the model group were given female and male cage mating pregnancy, then the control group and the pre-pregnancy stress induced PPD model group. The open field test was carried out on the female mice 3 weeks after delivery. Tail suspension test and sugar water preference test were used to evaluate the success of the model. The expression of DISC1 gene and protein was detected in hippocampus, serum estradiol and corticosterone levels were detected in orbital blood samples. Compared with the control group, the spontaneous activity of PPD model group did not change in the open field test, but the desperate immobility time in the tail suspension test increased significantly (P 0.001), and the sugar water preference decreased significantly. Compared with the control group, the hippocampal DISC1 gene t-8.915P0.001 and the protein were significantly decreased in the PPD model group. The expression of TG-5.004 was significantly up-regulated, and the expression of P0.01 was significantly up-regulated. Serum corticosterone and estradiol had no change (P 0.05). Conclusion chronic prepregnancy stress can induce postpartum depressive symptoms in Balb/c rats, and its pathogenesis may be related to the regulation of gene and protein expression of hippocampal DISC1.
