【摘要】：為了研究擬南芥基因AtHHR1(編碼一個含有RING finger結構域的E3連接酶)是否參與熱脅迫響應,構建了athhr1/AtHHR1互補轉基因株系.對突變體、互補株系及野生型進行熱脅迫處理,發(fā)現(xiàn)突變體的萌發(fā)率、葉綠素及脯氨酸含量高于野生型,而互補株系均低于野生型.通過real-time PCR定量檢測發(fā)現(xiàn),熱誘導后擬南芥熱信號通路中相關熱激蛋白基因在突變體中表達比野生型中高.研究結果初步表明AtHHR1基因在擬南芥的熱脅迫響應中起負調(diào)控作用.
[Abstract]:In order to study whether Arabidopsis thaliana gene AtHHR1 (encoding an E3 ligase containing RING finger domain) is involved in heat stress response, a complementary athhr1/AtHHR1 transgenic strain was constructed. It was found that the germination rate, chlorophyll and proline content of the mutant were higher than those of the wild type, but that of the complementary lines were lower than that of the wild type. The expression of heat shock protein gene in heat signal pathway of Arabidopsis thaliana was higher than that in wild type after heat induction. The results showed that AtHHR1 gene played a negative role in the heat stress response of Arabidopsis thaliana.
