冬小麥膨脹素基因TaEXPB8、TaEXPB10的克隆及低溫表達(dá)分析

【摘要】:以冬小麥強(qiáng)抗寒品種東農(nóng)冬麥1號(hào)為試驗(yàn)材料,從其根中克隆膨脹素基因TaEXPB8、TaEXPB10,并分析其在低溫條件下的表達(dá)情況,為研究膨脹素基因與植物抗寒性之間的關(guān)系奠定基礎(chǔ)。結(jié)果表明,在冬小麥強(qiáng)抗寒品種東農(nóng)冬麥1號(hào)根中克隆得到膨脹素基因TaEXPB8、TaEXPB10,二者分別編碼273、271個(gè)氨基酸,二者序列與Gen Bank上公布的序列同源性分別達(dá)到99.2%、99.6%。4℃、-10℃和-20℃的低溫脅迫處理均可誘導(dǎo)TaEXPB8、TaEXPB10基因在冬小麥強(qiáng)抗寒品種東農(nóng)冬麥1號(hào)根中表達(dá),尤其是TaEXPB10基因,其受誘導(dǎo)程度高于TaEXPB8;兩基因在低抗寒品種濟(jì)麥22號(hào)中總體上不受誘導(dǎo),甚至受到抑制。綜合比較發(fā)現(xiàn),4℃、-10℃和-20℃低溫脅迫下,TaEXPB8、TaEXPB10基因在強(qiáng)抗寒品種東農(nóng)冬麥1號(hào)中的表達(dá)量均明顯高于低抗寒品種濟(jì)麥22號(hào)。據(jù)此推測(cè)膨脹素基因TaEXPB8、TaEXPB10與冬小麥根系的低溫脅迫耐受能力具有重要的相關(guān)性,有助于進(jìn)一步深入了解冬小麥根系的抗寒機(jī)制。
[Abstract]:A strong cold resistant winter wheat variety Dongnong Dongmai 1 was used to clone the expandin gene TaEXPB8 and TaEXPB10 from its roots, and the expression of TaEXPB8 and TaEXPB10 at low temperature was analyzed. The results showed that the expandin gene TaEXPB8 and TaEXPB10 were cloned from the root of Dongnong Dongmai, a strong cold resistant winter wheat variety, and the two genes encoded 273,271 amino acids, respectively, in order to study the relationship between expandin gene and plant cold resistance. The sequence of TaEXPB8 and TaEXPB10 gene expressed in the root of Dongnongmai 1, a strong cold resistant winter wheat cultivar, could be induced by low temperature stress of 99.2 ~ 99.6C and at -20 鈩,
