【摘要】：薏苡Waxy基因是控制薏苡直鏈淀粉合成的關(guān)鍵基因,對(duì)其進(jìn)行生物信息學(xué)分析,為深入研究該基因的作用奠定基礎(chǔ)并為進(jìn)一步改良糯性新材料提供理論支撐,以薏苡的嫩莖為實(shí)驗(yàn)材料,采用RT-PCR方法克隆薏苡Waxy基因,使用NCBI、DNAMAN及ExPYSy等一系列在線軟件及工具,對(duì)薏苡Waxy基因的編碼區(qū),氨基酸序列及蛋白質(zhì)的結(jié)構(gòu)和功能進(jìn)行生物信息學(xué)分析,研究表明薏苡Waxy基因有13個(gè)內(nèi)含子,全長(zhǎng)1 845 bp,編碼由614個(gè)氨基酸,屬于糖基轉(zhuǎn)移酶超家族,薏苡與高粱親緣關(guān)系較近,同源性達(dá)93%;該蛋白其中α螺旋占35.18%,無(wú)規(guī)則卷曲所占比例33.22%,保守性較強(qiáng),通過(guò)對(duì)該蛋白保守性分析發(fā)現(xiàn):糖基轉(zhuǎn)移酶、糖原/淀粉合成酶和淀粉合成酶催化區(qū)3個(gè)保守性結(jié)構(gòu)域。
[Abstract]:The Waxy gene is Coix Coix amylose biosynthesis key genes, bioinformatics analysis of its, which laid the foundation for the further study of the gene function and provide theoretical support for further improvement of waxy materials, with Coix stems as experimental materials, RT-PCR method to clone Waxy gene by using Coix NCBI, DNAMAN. ExPYSy and a series of online software and tools of Coix Waxy gene encoding region, amino acid sequence and protein structure and function by bioinformatics analysis, studies show that Coix Waxy gene contains 13 introns, the total length of 1845 BP, encoding 614 amino acid, which belongs to the glycosyltransferase superfamily, is near the relationship between Coix and sorghum genetic, 93% homology of the protein; the alpha helix random coil accounted for 35.18%, the proportion of 33.22% of the protein by strong conservatism, conservative analysis: glycosyltransferase, sugar The 3 conserved domains of the original / starch synthetase and starch synthase catalyzed regions.
