【摘要】：為了闡明新疆油雞H9亞型流感病毒PB1基因的分子特征與進(jìn)化趨勢,本研究采用RT-PCR技術(shù)對新疆油雞分離株A/Chicken/Xinjiang Baicheng/1/2014(H9 N2)的PB1基因進(jìn)行了克隆、測序及分子進(jìn)化分析,將PB1基因的核苷酸序列和對應(yīng)的氨基酸序列與Genbank中已經(jīng)公布的參考序列進(jìn)行同源性比較。結(jié)果表明,新疆油雞分離株的PB1基因由2274個(gè)堿基組成,編碼757個(gè)氨基酸,與AIV A/chicken/Henan/89/2013(H7N9)的PB1基因處于同一分支,其核苷酸和氨基酸水平的同源性依次為99.2%和99.4%,新疆油雞分離株的PB1基因不屬于歐亞分支G1、A/CK/BJ/94和Y439的任何譜系。
[Abstract]:In order to elucidate the molecular characteristics and evolutionary trend of PB1 gene of Xinjiang oil chicken H9 subtype influenza virus, the PB1 gene of Xinjiang oil chicken isolate A / Chicken / Xinjiang Baicheng/1/2014(H9 N2 was cloned, sequenced and analyzed by RT-PCR. The nucleotide sequence and corresponding amino acid sequence of PB1 gene were compared with the reference sequence of Genbank. The results showed that the PB1 gene of Xinjiang oil chicken isolate was composed of 2274 bases and encoded 757 amino acids. The nucleotide and amino acid homology of PB1 gene of AIV A / chicken / Henan / 89 / 2013 H7N9) was 99.2% and 99.4, respectively. The PB1 gene of Xinjiang oil chicken isolate did not belong to any lineage of Eurasian branch G1Ar / CK / BJ94 and Y439.
