【摘要】：為了探索連梅提取液對濕熱瀉痢仔豬的抗氧化功能、胃腸調節(jié)酶及十二指腸凋亡基因表達的影響,試驗采用了人工模擬高濕高熱的環(huán)境,給仔豬灌服大腸桿菌輔助致使瀉痢,選擇瀉痢仔豬分為模型組、陽性藥物組和連梅提取液高、中、低劑量組,以正常環(huán)境下正常飼喂的仔豬作為對照,通過檢測仔豬血清抗氧化酶、胃腸調節(jié)酶、肝ATP酶以及十二指腸凋亡基因Caspase-3、-8、-9 mRNA轉錄變化以分析連梅提取液治療瀉痢仔豬的作用機制。結果顯示,濕熱瀉痢仔豬經連梅提取液進行治療后,血清T-SOD、GSH-Px活力升高,LDH、MDA含量減少,與模型組相比較差異極顯著(P0.01);乙酰膽堿酯酶、淀粉酶、肝Na+-K+-ATPase和Ca2+-Mg2+-ATPase活力較模型組顯著增強(P0.05);T3、T4和GH分泌量顯著升高(P0.05),GC分泌量顯著降低(P0.05);同時各藥物組仔豬十二指腸凋亡基因得到了不同程度的恢復,Caspase-8 mRNA表達量只有中劑量組較模型組極顯著降低至2.62倍(P0.01);Caspase-3mRNA表達中,高、中劑量組分別降至正常組的5.82、3.29倍,較模型組差異顯著(P0.05)或極顯著(P0.01);Caspase-9mRNA表達量均顯著降低(P0.05),且降低量與藥物濃度成正比。由此可知,連梅提取液可以通過增強濕熱瀉痢仔豬抗氧化損傷能力,改善胃腸道的消化吸收,促進生長激素的分泌和降低凋亡基因的表達而達到治療仔豬濕熱瀉痢。
[Abstract]:In order to explore the effects of Lianmei extract on the antioxidant function, gastrointestinal regulatory enzyme and duodenal apoptotic gene expression in piglets with damp-heat diarrhea, artificial simulation of high humidity and high fever was used in the experiment. The piglets were divided into three groups: the model group, the positive drug group and the high, middle and low dose groups. The piglets fed in normal environment were used as the control. Serum antioxidant enzymes, gastrointestinal regulatory enzymes, liver ATP enzymes and duodenal apoptotic gene Caspase-3 were detected in piglets. The transcriptional changes of -9 mRNA were used to analyze the mechanism of Lianmei extract in treating diarrhea piglets. The results showed that the serum T-SOD of damp-heat diarrhea piglets was treated with Lianmei extract. The activity of GSH-Px increased and the content of GSH-Px decreased, compared with the model group, the difference was very significant (P 0.01). The activities of acetylcholinesterase, amylase, liver Na K ATPase and Ca2 mg 2 ATPase were significantly increased compared with the model group. The secretion of T3, T4 and GH increased significantly in P0. 05 and P0. 05 and decreased significantly in P0. 05. At the same time, the apoptotic genes of duodenum were recovered to different degrees in the piglets of different drug groups. The expression of Caspase-8 mRNA in middle dose group was significantly lower than that in model group to 2.62 times (P 0.01). The expression of Caspase-3mRNA in the high and middle dose groups decreased to 5.82 ~ 3.29 times of the normal group, which was significantly higher than that of the model group (P 0.05) or extremely significant (P 0.01). The expression of Caspase-9mRNA was significantly lower than that of P0.05P, and the decrease was directly proportional to the concentration of the drug. The extract of Lianmei can improve the ability of anti-oxidation damage, improve the digestion and absorption of gastrointestinal tract, promote the secretion of growth hormone and decrease the expression of apoptotic gene in the piglets by enhancing the ability of anti-oxidative damage, promoting the secretion of growth hormone and reducing the expression of apoptotic gene.
