不同品種甜高粱SS基因表達與糖分積累關(guān)系的研究

【摘要】:甜高粱是新一代能源作物,是生產(chǎn)乙醇燃料的重要原料,甜高粱含糖量是決定乙醇產(chǎn)量的主要因素,提高甜高粱含糖量是當今甜高粱育種最重要的研究內(nèi)容之一。本實驗選取了國內(nèi)外10個甜高粱主栽品種,比較不同品種甜高粱不同時期的糖含量以及蔗糖合成酶和蔗糖磷酸合成酶的活性變化,利用實時熒光定量PCR技術(shù)比較4個時期、10個甜高粱品種之間SS基因表達的差異,了解甜高粱體內(nèi)糖分累積規(guī)律,主要結(jié)果如下:1、在不同的生長時期蔗糖含量和總糖含量變化規(guī)律比較一致,均呈逐漸上升趨勢,在成熟期時達到峰值。果糖和葡萄糖含量的變化趨勢與總糖的變化趨勢不同,果糖含量的變化整體上呈現(xiàn)升高、降低再增高的變化趨勢,葡萄糖含量總體上是呈現(xiàn)出增高、降低的變化趨勢。對甜高粱葉片中的糖含量進行相關(guān)性分析,結(jié)果得出,葉片中總糖含量與蔗糖含量呈顯著正相關(guān)(R=0.632,P0.01),總糖含量與果糖含量無相關(guān)性(R=0.227,P=0.159),總糖含量與葡萄糖含量呈負相關(guān)(R=-0.328,P0.05)。2、葉片中SS活性和SPS活性的變化趨勢大體相同,整體上呈現(xiàn)上升的趨勢,成熟期達到最大值,葉片中SS的酶活性大于SPS酶活性。在生長前期時,葉片中SS和SPS酶活性存在差別不大,在糖類積累的生長后期,SS和SPS酶活性才表現(xiàn)出明顯的差別。葉片中蔗糖含量與SS酶活性呈正相關(guān)(R=0.860,P0.05),與SPS酶活性也呈正相關(guān)(R=0.876,P0.05),說明SS和SPS是影響甜高粱蔗糖積累過程的主要的酶。在成熟期時品種LTR108的SS酶活性和SPS酶活性均為最大。3、通過對葉片中SS基因相對表達量定量分析可知,在成熟時期SS基因相對表達量最高的是LTR108這一品種,這樣的表達差異與蔗糖含量差異一致,說明甜高粱SS基因的表達量是影響蔗糖含量的因素,從而導致不同品種的甜高粱之間蔗糖含量的不同。
[Abstract]:Sweet sorghum is a new generation of energy crops, is an important raw material for the production of fuel ethanol, sugar content of sweet sorghum is the main factor to determine the ethanol yield, increase the sugar content of sweet sorghum is one of the most important research content in sweet sorghum breeding. This experiment selects 10 domestic and foreign sweet sorghum varieties, comparison of sugar content in different periods of different varieties of sweet sorghum and the changes of the activity of sucrose synthase and sucrose phosphate synthase, using real-time fluorescence quantitative PCR technology 4 times, the difference of SS gene expression between 10 varieties of sweet sorghum, sweet sorghum sugar accumulation in understanding, the main results are as follows: 1, compared with the variation in different growth periods of sucrose content and total sugar content were. Increased gradually, reached a peak at mature stage. The different trend of the content of fructose and glucose and total sugar, fructose content change The overall trend of increased, decreased and then increased, the content of glucose in general are showing increased, the trend of decrease. By correlation analysis of sugar content of sweet sorghum leaves in the leaves of total sugar content and sucrose content showed a significant positive correlation (R=0.632, P0.01), there was no correlation between total sugar content with the content of fructose (R=0.227, P=0.159), the total sugar content was negatively correlated with the content of glucose (R=-0.328, P0.05).2, the change trend of SS activity and SPS activity in the leaves of roughly the same, the overall upward trend, the maturity stage and reached the maximum enzyme activity in leaf of SS was greater than SPS in enzyme activity. The early growth stage, SS and SPS activity in leaves has little difference, carbohydrate accumulation in late growth stage, SS and SPS activity did not show significant differences. In the leaves of sucrose content and SS enzymatic activity was positively correlated (R=0.860, P0.05), and SPS activity. It was positively correlated (R=0.876, P0.05), SS and SPS are the main enzymes of sucrose accumulation. In the mature stage of SS enzyme activity and SPS activity of varieties of LTR108 were the highest in the leaves of SS.3, based on the analysis of relative expression quantity of SS gene, in the mature period of relative the expression of LTR108 was the highest of the varieties, consistent differences expressed this with sucrose content, indicating the expression of SS gene is the influence factors of sweet sorghum sucrose content, resulting in different varieties of sweet sorghum sucrose content is different.
