小干擾RNA沉默YKL-40基因?qū)ψ訉m內(nèi)膜癌HEC-1A細(xì)胞順鉑化療敏感度的影響

【摘要】:目的通過si RNA沉默YKL-40基因探討其對(duì)子宮內(nèi)膜癌HEC-1A細(xì)胞順鉑化療敏感度的影響。方法 si RNA基因干擾技術(shù)沉默YKL-40基因,將人子宮內(nèi)膜癌HEC-1A細(xì)胞分為三組:空白對(duì)照組、空載體組、si RNA實(shí)驗(yàn)組。q RT-PCR法檢測(cè)沉默效率。q RT-PCR法檢測(cè)順鉑處理子宮內(nèi)膜癌細(xì)胞對(duì)癌細(xì)胞中YKL-40基因的影響。MTT法和流式細(xì)胞術(shù)檢測(cè)靶細(xì)胞的增殖能力以及凋亡率。結(jié)果轉(zhuǎn)染組YKL-40m RNA的表達(dá)量顯著低于空白對(duì)照組和空載體組(P=0.036,P=0.005),經(jīng)過相同濃度的順鉑處理子宮內(nèi)膜癌后,YKL-40基因的表達(dá)量上調(diào)。轉(zhuǎn)染組的增殖能力低于空白對(duì)照組和空載體組(P0.05),而凋亡率高于空白對(duì)照組和空載體組(P=0.000,P=0.000)。結(jié)論 YKL-40基因沉默可以提高子宮內(nèi)膜癌HCE-1A細(xì)胞對(duì)順鉑的敏感度。YKL-40基因具有抗凋亡作用。
[Abstract]:Objective to investigate the effect of si RNA silencing YKL-40 gene on chemotherapeutic sensitivity of HEC-1A cells with endometrial carcinoma. RNA gene interference technique silenced YKL-40 gene. Human endometrial carcinoma HEC-1A cells were divided into three groups: blank control group and empty vector group. Si RNA experimental group. Q RT-PCR method to detect silencing efficiency. Q. RT-PCR assay to detect the effect of cisplatin treatment on YKL-40 gene in endometrial cancer cells. MTT assay and flow cytometry were used to detect the proliferation ability and apoptosis rate of target cells. L-40m. The expression of RNA was significantly lower than that of blank control group and empty vector group (. P0. 036. The expression of YKL-40 gene in endometrial carcinoma treated with the same concentration of cisplatin was up-regulated. The proliferation ability of transfected group was lower than that of blank control group and empty vector group (P0.05). The rate of apoptosis was higher than that of control group and empty carrier group. Conclusion YKL-40 gene silencing can increase the sensitivity of HCE-1A cells to cisplatin. YKL-40 gene has anti-apoptosis effect.
