急性脅迫對團(tuán)頭魴鐵穩(wěn)態(tài)及其相關(guān)基因表達(dá)的影響

【摘要】:為研究急性應(yīng)激反應(yīng)對魚體鐵含量及鐵穩(wěn)態(tài)相關(guān)基因的影響,以團(tuán)頭魴為研究對象,通過腹腔注射皮質(zhì)醇來模擬急性應(yīng)激,在注射后不同時(shí)間點(diǎn)對團(tuán)頭魴的血液、腸和肝臟進(jìn)行取樣,采用分光光度法測定血清和肝臟中的鐵含量,采用實(shí)時(shí)熒光定量PCR對鐵穩(wěn)態(tài)相關(guān)基因的表達(dá)量進(jìn)行檢測。結(jié)果顯示,皮質(zhì)醇注射后血漿皮質(zhì)醇顯著升高,血漿中鐵含量在4、8、10和12 h顯著高于對照組,而肝臟中的鐵含量在4、8、10和12 h時(shí)顯著低于對照組。團(tuán)頭魴肝臟中鐵調(diào)素(hep)基因表達(dá)量顯著增加,并分別在8和10 h達(dá)最大值,隨后有所下降。腸道和肝臟中轉(zhuǎn)鐵蛋白基因(Tf)的表達(dá)量均呈先升高后下降的趨勢。研究表明,急性應(yīng)激下機(jī)體細(xì)胞內(nèi)儲存鐵大量釋放到體液中,造成病原感染和體內(nèi)增殖風(fēng)險(xiǎn)增加;hep及Tf上調(diào)表達(dá)降低細(xì)胞內(nèi)鐵釋放和促進(jìn)體液鐵向胞內(nèi)蓄積,在魚體內(nèi)鐵穩(wěn)態(tài)的調(diào)控和固有免疫反應(yīng)中發(fā)揮著重要的作用。
[Abstract]:In order to study the effect of acute stress on iron content and iron homeostasis genes in fish, acute stress was simulated by intraperitoneal injection of cortisol. Blood, intestine and liver were sampled at different time points after injection. The iron content in serum and liver was determined by spectrophotometry. The expression of iron-homeostasis related genes was detected by real-time fluorescence quantitative PCR. The results showed that the plasma cortisol level was significantly increased after cortisol injection, and the iron content in plasma was 4 ~ 8%. At 10 h and 12 h, the iron content in liver was significantly lower than that in control group. The expression of transferrin gene Tfin in intestine and liver increased first and then decreased. Under acute stress, iron stored in cells was released into body fluid, which resulted in increased risk of pathogenic infection and proliferation in vivo. The up-regulated expression of hep and TF plays an important role in the regulation of iron homeostasis and inherent immune response in fish.
