【摘要】：化學(xué)感受蛋白(chemosensory protein,CSPs)在昆蟲的免疫反應(yīng)、生長(zhǎng)發(fā)育及信號(hào)傳遞過程中起重要作用。以耐高溫家蠶品種為材料,研究高溫飼養(yǎng)環(huán)境下,作為家蠶物理屏障及感覺器官的表皮中化學(xué)感受蛋白基因的表達(dá)變化。依據(jù)從家蠶基因組數(shù)據(jù)庫獲得的15個(gè)家蠶CSPs基因(Bm CSPs)的序列設(shè)計(jì)引物,通過RT-PCR檢測(cè)正常飼育溫度(28℃±0.5℃)下,Bm CSP1、Bm CSP4、Bm CSP6、Bm CSP7、Bm CSP8、Bm CSP9、Bm CSP11和Bm CSP15等8個(gè)基因在家蠶5齡第2天幼蟲的表皮中有表達(dá);通過半定量RT-PCR和qRT-PCR檢測(cè)在高溫(35℃±0.5℃)飼育環(huán)境下,5齡第2天幼蟲表皮中Bm CSP8和Bm CSP15的表達(dá)水平顯著上調(diào),其他6個(gè)Bm CSPs基因的表達(dá)水平未見顯著變化。初步推測(cè)Bm CSP8和Bm CSP15可能與家蠶幼蟲對(duì)高溫環(huán)境的應(yīng)激反應(yīng)有關(guān)。
