【摘要】:由落葉松-楊柵銹菌引起的楊樹(shù)葉銹病分布廣泛,危害嚴(yán)重。為探究楊樹(shù)C14族R2R3-MYB基因在楊樹(shù)抗銹菌過(guò)敏性反應(yīng)中的調(diào)控作用,對(duì)接種了落葉松-楊柵銹菌E4強(qiáng)致病性生理小種的2種楊樹(shù)葉片進(jìn)行癥狀觀察、感病指數(shù)判定并對(duì)2種楊樹(shù)C14族共6條R2R3-MYB基因表達(dá)量變化情況進(jìn)行分析。接種7 dpi后,弱感病性樹(shù)種雜交楊葉片產(chǎn)生的夏孢子堆數(shù)量顯著少于感病樹(shù)種歐美楊。雜交楊葉片4 dpi開(kāi)始出現(xiàn)過(guò)敏性反應(yīng)癥狀,歐美楊未觀察到類(lèi)似癥狀。表明過(guò)敏性反應(yīng)可有效阻止銹菌夏孢子堆形成,降低楊樹(shù)感病性。雜交楊與歐美楊R2R3-MYB基因表達(dá)量變化趨勢(shì)相反,預(yù)測(cè)楊樹(shù)C14族R2R3-MYB基因?yàn)闂顦?shù)與病原互作過(guò)程中調(diào)控過(guò)敏性反應(yīng)的正向調(diào)控因子。本研究為進(jìn)一步系統(tǒng)研究楊樹(shù)R2R3-MYB基因與過(guò)敏性反應(yīng)的調(diào)控機(jī)理及楊樹(shù)抗病育種工作提供理論依據(jù)。
[Abstract]:Poplar leaf rust caused by Larix gmelinifera was widely distributed and seriously damaged. In order to investigate the role of R2R3-MYB gene in the allergic response of poplar to rust resistance, R2R3-MYB gene of poplar C14 was studied. The leaf symptoms of two species of poplar inoculated with AE4 strong pathogenicity of Larix gmelinifera were observed. Susceptibility index was used to determine and analyze the expression of 6 R2R3-MYB genes in two species of poplar C14. After inoculation for 7 dpi, the expression of R2R3-MYB gene was analyzed. The number of summer spores produced in poplar leaves of weakly susceptible poplar was significantly less than that of susceptible poplar Euramerican poplar. The anaphylaxis symptoms began to appear in the leaves of hybrid poplar for 4 dpi. No similar symptoms were observed in Euramerican poplar, which indicated that anaphylaxis reaction could effectively prevent the formation of summer spores and reduce the susceptibility of poplar. The change trend of R2R3-MYB gene expression in hybrid poplar and Euramerican poplar was opposite. It is predicted that the R2R3-MYB gene of poplar C14 is a positive regulator of allergic reaction in poplar interaction with pathogen. This study is intended to further systematically study the relationship between R2R3-MYB gene and allergies in poplar. The regulation mechanism of sex response and the breeding of poplar disease resistance provide theoretical basis.
