【摘要】：培養(yǎng)心肌細(xì)胞是生物醫(yī)學(xué)研究領(lǐng)域一項(xiàng)新實(shí)驗(yàn)技術(shù),在心血管病理生理學(xué)研究中起著相當(dāng)重要的作用。本學(xué)科自1998年為研究生開設(shè)《心肌細(xì)胞分離與培養(yǎng)》選修課,目的是使學(xué)生掌握心肌細(xì)胞分離培養(yǎng)的基本原理和方法并應(yīng)用于科學(xué)研究中。講義編寫和講授內(nèi)容以原代細(xì)胞培養(yǎng)為線索,以大鼠心肌細(xì)胞分離和培養(yǎng)為實(shí)例,比較詳盡地講解和演示了實(shí)驗(yàn)的全過程。主要內(nèi)容:(1)分離:①未成熟心肌細(xì)胞:準(zhǔn)備→取材→酶解→離心→制備細(xì)胞懸液;②成熟心肌細(xì)胞:準(zhǔn)備Langendroff裝置→摘取心臟和插管→灌流→酶解→離心復(fù)鈣。(2)培養(yǎng):包括貼壁培養(yǎng)和懸浮培養(yǎng)。重點(diǎn)講授純化心肌細(xì)胞、酶選擇、消化程度把握、各部位心肌細(xì)胞培養(yǎng)、2種培養(yǎng)方法差異與表達(dá)的特性、生長狀態(tài)等。(3)建立相關(guān)疾病模型:如心肌肥厚、心衰等。(4)應(yīng)用:包括心肌細(xì)胞超微結(jié)構(gòu)、細(xì)胞內(nèi)外離子轉(zhuǎn)運(yùn)通道、細(xì)胞凋亡、受體、蛋白質(zhì)組學(xué)、藥物學(xué)研究等。該方法稍加改良亦可用于雞、小鼠、兔、狗等動物及人類胎兒和成人手術(shù)獲得的心肌碎片的心肌細(xì)胞培養(yǎng)。通過多年探索,已基本完善了教學(xué)內(nèi)容,提高了教學(xué)質(zhì)量和教學(xué)水平。
[Abstract]:Cardiomyocyte culture is a new experimental technique in biomedical research, which plays an important role in cardiovascular pathophysiology. The course of cardiomyocyte separation and culture has been offered to graduate students since 1998. The purpose of this course is to make students master the basic principles and methods of cardiomyocyte separation and culture and apply them to scientific research. Taking primary cell culture as clue and rat cardiomyocyte isolation and culture as an example, the whole process of the experiment is explained and demonstrated in detail. The main contents were as follows: (1) isolation: 1immature cardiomyocytes: preparation of cell suspensions by centrifugation; (2) mature cardiomyocytes: preparation of Langendroff device for the extraction of cardiac cells and the centrifugation of pericardial pericardium. (2) culture, including adherent culture and suspension culture. Emphasis is placed on purified cardiomyocytes, enzyme selection, digestion degree assurance, cardiomyocyte culture in different parts, differences and expression characteristics of two culture methods, growth state, etc. (3) establishing models of related diseases, such as myocardial hypertrophy, and so on. (4) applications include ultrastructure of cardiomyocytes, intracellular and extracellular ion transport channels, apoptosis, receptors, proteomics, pharmacology, and so on. The method can also be used to culture cardiac muscle cells from chickens, mice, rabbits, dogs, human fetuses and adults. Through many years of exploration, the teaching content has been basically improved, and the teaching quality and teaching level have been improved.
