B7-H3分子介導(dǎo)的結(jié)直腸癌細(xì)胞耐藥機(jī)制的探究

結(jié)直腸癌(CRC)是臨床常見的癌癥類型之一,每年大約有120萬新結(jié)直腸癌確診病例,而每年死于結(jié)直腸癌患者人數(shù)約為60萬。化療藥物耐藥的出現(xiàn)是結(jié)直腸癌高死亡率的一個重要原因,了解導(dǎo)致個體化療藥物耐藥的機(jī)制有助于確定新的藥物靶點和藥物研發(fā),從而改善癌癥治療。本課題組前期發(fā)現(xiàn)B7-H3能夠促進(jìn)結(jié)直腸癌細(xì)胞的遷移和侵襲。因為腫瘤轉(zhuǎn)移和化療耐藥具有密切聯(lián)系,在本課題中,我們以B7-H3為靶點探究其與結(jié)直腸癌細(xì)胞耐藥的關(guān)系。首先,我們分析了共刺激分子B7-H3在HCT-116、HCT-8、HT29、SW480、SW620、RKO、LOVO細(xì)胞中的表達(dá)。Westernblot結(jié)果發(fā)現(xiàn),相對于SW480、SW620、RKO、LOVO細(xì)胞,B7-H3在HCT-8、HT29、HCT-116細(xì)胞中有更高的表達(dá)。我們發(fā)現(xiàn)B7-H3分子在HCT-8/5-Fu以及HCT-116/L-OHP兩組耐藥細(xì)胞中的表達(dá)均高于其野生細(xì)胞(HCT-8,HCT-116)。為了評估B7-H3分子對CRC細(xì)胞藥物敏感性的影響,我們使用特定的siRNA沉默了HCT-116和HCT-8細(xì)胞中B7-H3的表達(dá)。藥物敏感性實驗結(jié)果顯示HCT...

第二章 B7-H3 表達(dá)對結(jié)直腸癌細(xì)胞對奧沙利鉑敏感性的影響3 表達(dá)對結(jié)直腸癌細(xì)胞細(xì)胞凋亡的影響驗中我們將敲減 B7-H3 的 HCT-8/RNAi 細(xì)胞與其對照組 HCT-8/Scra度為 30μg/mL 的奧沙利鉑作用,經(jīng) Annexin/PI 染色,結(jié)果如圖 2-胞的細(xì)胞凋亡率約為 46.5%而 HCT-8/Scrambled 細(xì)胞的細(xì)胞凋亡率實驗驗證中我們將 SW480-B7H3 細(xì)胞及其對照組細(xì)胞與 20μg/mL nnexin/PI 染色,實驗結(jié)果如圖 2-3B 所示,SW480-B7H3 細(xì)胞的對照組細(xì)胞 SW480-NC 細(xì)胞的凋亡率約為 43.1%,說明 SW480-B-NC 相比抗凋亡的能力更強(qiáng)。通過以上實驗結(jié)果說明,B7-H3 分子腸癌細(xì)胞的抗凋亡能力。AHCT-8/Scrambled HCT-8/RNAi
