PI3K/mTOR抑制劑NVP-BEZ235抑制甲狀腺癌細胞增殖并促進細胞凋亡的作用研究

目的:研究PI3K/mTOR信號通路抑制劑NVP-BEZ235體外抑制甲狀腺癌細胞的增殖,并誘導(dǎo)其凋亡的作用及分子機制,為甲狀腺癌的臨床治療提供新的分子靶點。方法:體外培養(yǎng)甲狀腺癌SW579細胞,采用MTT檢測抑制劑NVP-BEZ235對甲狀腺癌SW579細胞的抑制作用,繪制SW579生存曲線,篩選藥物濃度。SW579細胞中加入100nM、2501μM的BEZ235藥物,對照組加入等體積的DMSO,作用48h后,提取蛋白,應(yīng)用Western blot法檢測甲狀腺癌SW579細胞中細胞周期蛋白Cyclin A、Cyclin B1和Cyclin D1的變化情況,檢測BEZ235對SW579細胞周期的抑制作用。再次應(yīng)用Western blot法檢測PI3K/mTOR信號通路蛋白和凋亡蛋白的變化情況。結(jié)果:1. NVP-BEZ235可抑制SW579細胞的增殖。MTT實驗結(jié)果表明,用BEZ235阻斷PI3K/mTOR信號遙路可抑制甲狀腺癌細胞SW579的增殖;Western blot結(jié)果表明,甲狀腺癌SW579細胞中Cyclin A、Cyclin B1和Cyclin D1蛋白的表達水平均隨BEZ...
