ECRG4表達(dá)與胃癌臨床病理學(xué)關(guān)系及相關(guān)機(jī)制的研究

目的:近年來惡性腫瘤的發(fā)生率和死亡率呈上升趨勢,嚴(yán)重危害著人類健康。胃癌是我國、日本、韓國等東亞國家高發(fā)惡性腫瘤之一,胃癌的發(fā)生發(fā)展是個人、環(huán)境及其他多種因素、多階段共同作用的結(jié)果,包括遺傳學(xué),表遺傳學(xué)及環(huán)境因素等。表遺傳學(xué)事件成為近些年研究的熱點,通過影響基因轉(zhuǎn)錄活性抑制基因表達(dá),卻不涉及DNA序列的改變。表遺傳學(xué)修飾包括DNA甲基化,組蛋白修飾,染色體重塑,RNA干擾等,其中DNA甲基化和組蛋白修飾兩種表遺傳學(xué)現(xiàn)象研究最為廣泛。腫瘤抑制基因失活包括基因缺失,基因突變和DNA異常甲基化三種機(jī)制。研究顯示胃癌發(fā)生、發(fā)展和形成過程中腫瘤抑制基因發(fā)生甲基化頻繁出現(xiàn)。食管癌相關(guān)基因4(Esophageal Cancer Relaetd Gene4,ECRG4),是98年中國醫(yī)學(xué)科學(xué)院Bi等利用m RNA差異顯示技術(shù)從食管癌組織中分離得到的。該基因位于人染色體2q14.1-14.3,有4個外顯子,全長12,500bp,其開放閱讀框架為444bp,編碼148個氨基酸多肽鏈,蛋白產(chǎn)物分子量為17k Da,在該基因的核心啟動子區(qū)、第一外顯子和第一內(nèi)含子5′端的大約1kb的范圍內(nèi)分布有多個Cp G島,...
