熱休克蛋白70在Bcr/Ab1細(xì)胞中抑制凋亡誘導(dǎo)因子的促凋亡作用

bcr/abl融合基因是慢性粒細(xì)胞白血。–hronic myloid leukemia, CML)的分子標(biāo)志,該基因表達(dá)具有組成型激活的酪氨酸激酶活性的Bcr/Abl融合蛋白,能夠異;罨疨I3K-AKT、Ras和STAT5等一系列信號(hào)通路,使血細(xì)胞惡性增殖和凋亡受阻。除此以外,Bcr/Abl融合蛋白表達(dá)的細(xì)胞中還能檢測(cè)到熱休克蛋白70(Hsp-70)表達(dá)水平增高,Hsp-70作為細(xì)胞應(yīng)激時(shí)的保護(hù)蛋白,近年來(lái)在腫瘤的發(fā)生發(fā)展過(guò)程中受到廣泛關(guān)注。Hsp-70可抑制Bax的線粒體定位,從而抑制線粒體通路的細(xì)胞凋亡;也可在線粒體下游抑制Apaf-1對(duì)caspase-9的募集作用,從而阻斷caspase凋亡通路的活化。此外,Hsp-70還能抑制非caspase凋亡通路。凋亡誘導(dǎo)因子(Apoptosis-inducing factor,AIF)是非caspase凋亡途徑的重要分子,生理?xiàng)l件下,AIF表達(dá)后依賴其N(xiāo)端1-120的線粒體定位信號(hào)(MLS)定位于線粒體,輔助呼吸鏈的電子傳遞。凋亡發(fā)生時(shí),AIF易位至細(xì)胞核中,誘發(fā)基因組DNA斷裂,促進(jìn)細(xì)胞凋亡。在MEF細(xì)胞中,外源性過(guò)表達(dá)Hsp-70...
