目的:研究Twist2在前列腺癌中的表達及其與腫瘤惡性程度間的內(nèi)在聯(lián)系。探索雄激素/AR與Twist2在雄激素依賴與雄激素非依賴前列腺癌細胞中的相互調(diào)控機制。揭示雄激素/AR-Twist2軸在前列腺癌惡性增殖、侵襲及EMT中的作用。材料與方法:收集上海交通大學醫(yī)學院附屬仁濟醫(yī)院泌尿科自2007年11月²014年4月間前列腺癌根治術(shù)標本93例,另取正常前列腺組織14例。免疫組化法檢測前列腺腫瘤組織與正常前列腺組織石蠟標本中Twist2的表達。在雄激素/AR與Twist2在前列腺癌中的相互調(diào)控作用研究中,先分別以0.1nmol/l、1nmol/l及10nmol/l濃度梯度的DHT處理LNCAP細胞系及PC-3細胞系,而后分別以RT-PCR及Western Blot分析Twist2的m RNA及蛋白表達情況。接著,使用比卡魯胺預處理LNCAP及PC-3細胞系,功能性抑制雄激素/AR軸,而后加入1nmol/l DHT,以RT-PCR檢測LNCAP及PC-3細胞系Twist 2的m RNA表達。進而以Western Blot測定LNCAP細胞系使用比卡魯胺功能性抑制雄激素/...

正常前列腺組織與前列腺癌組織Twist2免疫組化染色進一步根據(jù)D’Amico前列腺癌危險程度分級，將腫瘤組織分為高危組

：Gleason≤6、Gleason 7 及 Gleason≥8 前列腺腫瘤組織 Twist2 表ason≤6 的低危前列腺癌組，Twist2 表達陰性占 6.25%，弱陽性占.73%，強陽性占 0%；在 Gleason 7 的中危前列腺癌組，Twist2 弱陽性占 25%，陽性占 68.33%，強陽性占 1.67%；在 Gleason≥組，Twist2 表達陰性占 11.76%，弱陽性占 5.88%，陽性占 174.71%。（圖 1-3）
