G蛋白偶聯(lián)受體激酶2調(diào)控M2巨噬細胞β2腎上腺素受體信號在肝細胞癌中的作用

原發(fā)性肝細胞癌(hepatocellular carcinoma,HCC)約占原發(fā)性肝癌的90%,是一種高度惡性腫瘤,全球腫瘤相關(guān)死因第二位。由于HCC的病因尚未完全明確,并且缺乏特效的藥物與治療手段,HCC患者復(fù)發(fā)率高,預(yù)后較差。因此,關(guān)于HCC的新治療方法迫切需要。腫瘤微環(huán)境是指腫瘤發(fā)生發(fā)展過程中所處的局部生物環(huán)境,主要包括免疫細胞、新生血管細胞、腫瘤相關(guān)成纖維細胞以及一些細胞外基質(zhì)等。巨噬細胞為單核細胞譜系的單核吞噬白細胞,根據(jù)功能不同分為兩型,即M1和M2型巨噬細胞。腫瘤微環(huán)境中浸潤的巨噬細胞稱為腫瘤相關(guān)巨噬細胞(tumor associated macrophages,TAM),以M2型為主。研究表明,TAM與腫瘤進展相關(guān)。肝臟內(nèi)分布大量的自主神經(jīng),包括交感神經(jīng)和副交感神經(jīng)。交感神經(jīng)系統(tǒng)激活并釋放去甲腎上腺素和腎上腺素,能夠激活腫瘤細胞或宿主基質(zhì)細胞的β腎上腺素能受體(β-adrenergic receptor,β-AR),促進多種實體腫瘤的生長和轉(zhuǎn)移。β2-AR是β-AR的亞型,屬于G蛋白偶聯(lián)受體(G protein-coupled receptor,GPCR),具有七個跨...
