miR-21在肝癌中的表達(dá)及其與PTEN的關(guān)系

【摘要】:目的:探討miR-21在肝癌中的表達(dá)及其與PTEN的關(guān)系。方法:用RT-PCR檢測(cè)119例肝癌組織與癌旁組織中miR-21與PTEN的表達(dá)情況;雙報(bào)告基因?qū)嶒?yàn)檢測(cè)肝癌細(xì)胞中miR-21對(duì)PTEN轉(zhuǎn)錄水平的影響;分析miR-21表達(dá)與肝癌患者臨床病理因素的關(guān)系;在肝癌細(xì)胞中分別過(guò)表達(dá)及抑制miR-21的表達(dá)之后,檢測(cè)細(xì)胞增殖與PTEN及其下游通路蛋白的表達(dá)。結(jié)果:肝癌組織中miR-21表達(dá)明顯高于癌旁組織,而PTEN則相反(均P0.05);miR-21與PTEN的表達(dá)具有一定相關(guān)性(r2=0.6767,P0.05);miR-21可并抑制PTEN轉(zhuǎn)錄活性;miR-21的表達(dá)與肝癌患者AFP水平及TNM分期有關(guān)(均P0.05);肝癌細(xì)胞過(guò)表達(dá)miR-21后,肝癌細(xì)胞增殖明顯增強(qiáng),PTEN表達(dá)明顯調(diào),并影響其下游蛋白的表達(dá),而抑制miR-21則相反(均P0.05)。結(jié)論:miR-21在肝癌中表達(dá)升高,且miR-21可以通過(guò)下調(diào)PTEN表達(dá)進(jìn)一步促進(jìn)肝癌細(xì)胞的增殖。

Methods: the expression of miR-21 and PTEN in 119 HCC tissues and paracancerous tissues was detected by RT-PCR, the effect of miR-21 on PTEN transcription in HCC cells was detected by double reporter gene assay, the relationship between miR-21 expression and clinicopathological factors was analyzed, and the expression of PTEN and its downstream pathway proteins were detected after overexpression and inhibition of miR-21 expression in HCC cells.
