【摘要】：目的通過調(diào)節(jié)肺癌A549細(xì)胞中沉默信息調(diào)節(jié)因子1(SIRT1)的表達(dá),探討SIRT1對肺癌細(xì)胞增殖和轉(zhuǎn)移的影響及其分子機(jī)制。方法培養(yǎng)肺癌A549細(xì)胞,用重組腺病毒感染A549細(xì)胞上調(diào)/下調(diào)SIRT1的表達(dá);通過MTT實驗、克隆形成實驗、劃痕實驗和Transwell遷移實驗檢測上調(diào)/下調(diào)SIRT1表達(dá)對肺癌A549細(xì)胞增殖和轉(zhuǎn)移的影響;干擾SIRT1表達(dá)后用Western blot檢測SIRT1對肺癌A549細(xì)胞中上皮細(xì)胞向間質(zhì)細(xì)胞轉(zhuǎn)化(EMT)相關(guān)蛋白標(biāo)記物ZO-1、β-連環(huán)蛋白(β-catenin)、Snail和ZEB1表達(dá)的影響。結(jié)果上調(diào)/下調(diào)SIRT1表達(dá)對肺癌A549細(xì)胞增殖無明顯影響,但是可以促進(jìn)/抑制肺癌A549細(xì)胞遷移;下調(diào)SIRT1表達(dá)后EMT相關(guān)上皮性標(biāo)記蛋白ZO-1表達(dá)增多,間質(zhì)性標(biāo)記蛋白β-catenin、Snail和ZEB1表達(dá)減少。結(jié)論 SIRT1可能通過影響EMT相關(guān)標(biāo)記蛋白的表達(dá)促進(jìn)肺癌A549細(xì)胞遷移。
[Abstract]:Objective to investigate the effect of SIRT1 on proliferation and metastasis of lung cancer cells and its molecular mechanism by regulating the expression of silencing information regulator 1 (SIRT1) in lung cancer cell line A549. Methods Lung cancer A549 cells were cultured and infected with recombinant adenoviruses to up-regulate / down-regulate the expression of SIRT1, and the effects of up-regulation / down-regulation of SIRT1 expression on the proliferation and metastasis of lung cancer A549 cells were detected by MTT assay, clone formation test, scratch test and Transwell migration assay. After interfering with SIRT1 expression, the effect of SIRT1 on the expression of (EMT) related protein markers ZO-1, 尾-catenin), Snail and ZEB1 from epithelial cells to stroma cells in lung cancer A549 cells was detected by Western blot. Results the up-regulation / down-regulation of SIRT1 expression had no significant effect on the proliferation of lung cancer cell line A549, but it could promote / inhibit the migration of lung cancer cell line A549. After down-regulating the expression of SIRT1, the expression of EMT-related epithelial marker protein ZO-1 increased and the expression of interstitial marker protein 尾-catenin,Snail and ZEB1 decreased. Conclusion SIRT1 may promote the migration of lung cancer cell line A549 by affecting the expression of EMT-related marker protein.
