EGFR突變豐度的檢測及其臨床意義

【摘要】:具有表皮生長因子受體(epidermal growth factor receptor,EGFR)敏感突變的非小細胞肺癌患者對酪氨酸激酶抑制劑(tyrosine kinase inhibitors,TKIs)具有良好反應(yīng),但是即使存在相同突變,不同患者對TKIs的反應(yīng)也不一致。推測突變分子含量的不同是TKIs反應(yīng)不一的部分原因,具有更高EGFR突變"豐度"的腫瘤患者可能從TKIs中獲益更多。EGFR突變可根據(jù)檢測方法敏感性的不同,定量檢測突變分子比例以及突變蛋白表達等進行半定量或定量檢測。EGFR突變豐度可能有助于反映腫瘤異質(zhì)性,評估疾病進程,預(yù)測TKIs藥物敏感性,早期發(fā)現(xiàn)耐藥,具有重要的臨床意義。
[Abstract]:Non-small cell lung cancer patients with epidermal growth factor receptor (epidermal growth factor receptor,EGFR) sensitive mutation responded well to tyrosine kinase inhibitor (tyrosine kinase inhibitors,TKIs), but even if there was the same mutation, the response of different patients to TKIs was not the same. It is speculated that the difference of mutation molecular content is part of the reason for the different TKIs response, and tumor patients with higher EGFR mutation "abundance" may benefit more from TKIs. EGFR mutation can be semi-quantitatively or quantitatively detected according to the sensitivity of detection methods. EGFR mutation abundance may be helpful to reflect tumor heterogeneity, evaluate disease process and predict TKIs drug sensitivity. Early detection of drug resistance has important clinical significance.
