胃癌細(xì)胞促進(jìn)網(wǎng)膜脂肪干細(xì)胞分化的機(jī)制研究

【摘要】:目的:本實(shí)驗(yàn)主要研究在胃癌條件培養(yǎng)基(conditioned medium,CM)誘導(dǎo)下網(wǎng)膜脂肪干細(xì)胞(omental-adipose stromal cells,O-ASCs)是否能分化為癌相關(guān)成纖維細(xì)胞(carcinoma-associated fibroblasts,CAFs),及ERK信號(hào)通路在其中的作用。方法:通過誘導(dǎo)分化成骨、成脂及流式細(xì)胞鑒定O-ASCs,將O-ASCs與MGC803和SGC7901 CM共培養(yǎng),通過RT-PCR和Western-blot檢測O-ASCs細(xì)胞CAFs標(biāo)志物α-SMA、FSP-1、vimentin,旁分泌因子VEGFA、TGFβ-1、FAP、SDF-1的表達(dá)水平。將O-ASCs分為對(duì)照組,SGC7901-CM實(shí)驗(yàn)組,SGC7901-CM+U0126處理組,12 h后收集細(xì)胞。Western blot檢測O-ASCs細(xì)胞CAFs標(biāo)志物α-SMA、FSP-1及ERK1/2、p-ERK1/2的表達(dá)水平。結(jié)果:經(jīng)鑒定原代培養(yǎng)出的細(xì)胞為O-ASCs,在SGC7901 CM和MGC803 CM作用下,CAFs標(biāo)志物α-SMA、FSP-1、vimentin及旁分泌因子SDF-1、VEGFA、TGFβ-1、FAP表達(dá)均有明顯增加(P0.05)。與對(duì)照組比較SGC7901-CM組α-SMA、FSP-1、p-ERK1/2表達(dá)明顯增加(P0.05),ERK表達(dá)未見明顯變化(P0.05)。SGC7901-CM+U0126組與SGC7901-CM組比較,α-SMA、FSP-1及p-ERK1/2的蛋白表達(dá)水平明顯降低(P0.05),ERK表達(dá)變化無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:O-ASCs通過分化為CAFs及旁分泌作用參與胃癌腹膜轉(zhuǎn)移,ERK信號(hào)通路在該過程中發(fā)揮了重要作用。
