【摘要】：目的:探討不同劑量的維生素C(抗壞血酸,Vitamin C,VC)對(duì)奧沙利鉑(L-OHP)在人胃癌裸鼠模型中抗腫瘤療效的影響。方法:建立人胃癌細(xì)胞(SGC-7901)裸鼠皮下移植瘤模型,按照隨機(jī)數(shù)字表法分為8組,每組10只。A組腹腔注射5%葡萄糖注射液;B組腹腔注射L-OHP 6 mg/kg;C、D、E組分別腹腔注射低、中、高劑量VC 100、200、400 mg/kg;F、G、H組分別腹腔注射L-OHP 6 mg/kg及低、中、高劑量VC 100、200、400 mg/kg,各組葡萄糖、VC每天給藥1次,L-OHP每3天給藥1次,連續(xù)用藥15天。觀察裸鼠生活狀態(tài),測(cè)裸鼠體質(zhì)量、瘤體積,計(jì)算抑瘤率,繪制移植瘤生長(zhǎng)曲線;停藥后殺死動(dòng)物,檢測(cè)血常規(guī)、肝、腎功能,行腫瘤、肝病理組織學(xué)檢查。結(jié)果:VC與L-OHP之間存在交互作用(P0.01),呈拮抗性,VC的濃度越高,對(duì)L-OHP抑制腫瘤生長(zhǎng)的拮抗作用越強(qiáng)。C、D、E組未表現(xiàn)出抑制腫瘤生長(zhǎng)的作用(P0.05)。B、F、G、H組表現(xiàn)出抑制腫瘤生長(zhǎng)的作用(P0.01),且隨VC濃度增高,抑瘤率下降。結(jié)論:不同劑量VC均未表現(xiàn)出抑制胃癌細(xì)胞生長(zhǎng)的作用;VC對(duì)L-OHP的化療療效有拮抗作用,且呈劑量依賴性。
[Abstract]:Aim: to investigate the effect of different doses of vitamin C (ascorbic acid, Vitamin C, VC) on the antitumor effect of oxaliplatin (L-OHP) in nude mice with human gastric cancer. Methods: the subcutaneous transplantation tumor model of human gastric cancer cell (SGC-7901) in nude mice was established and divided into 8 groups with 10 mice in each group according to the random digital table method. 5% glucose injection was injected intraperitoneally in group A and low, middle and high dose VC 100200400 mg/kg; were injected intraperitoneally in group B and group B with L-OHP 6 mg/kg;C,D,E, respectively. F, G, H group were intraperitoneally injected with L-OHP 6 mg/kg and low, medium and high dose VC 100200400 mg/kg, groups, VC was given once a day, L-OHP was given once every 3 days for 15 days. The living state of nude mice was observed, the body mass and tumor volume of nude mice were measured, the tumor inhibition rate was calculated, and the growth curve of transplanted tumor was drawn. The animals were killed after drug withdrawal, the blood routine, liver and kidney function, tumor and liver pathology were examined. Results: there was interaction between VC and L-OHP (P 0.01). The higher the concentration of VC, the stronger the antagonistic effect on L-OHP inhibition of tumor growth. C, D, E group showed no inhibitory effect on tumor growth (P 0.05). B, F, G, H group showed inhibitory effect on tumor growth (P 0.01), and the tumor inhibition rate decreased with the increase of VC concentration. Conclusion: different doses of VC have no inhibitory effect on the growth of gastric cancer cells, and VC has antagonistic effect on the chemotherapy effect of L-OHP in a dose-dependent manner.
