【摘要】：目的:探討木姜子乙醇提取物(Litsea pungens ethanol extract,LPEE)對肝癌細胞侵襲轉移能力的影響及其相關機制。方法:MTT法測定LPEE對肝癌細胞株HepG2和QGY-7703的IC50;transwell遷移實驗和matrigel侵襲實驗檢測LPEE對HepG2和QGY-7703細胞遷移和侵襲能力的影響;qRT-PCR和Western blot檢測LPEE對各組細胞中E-cadherin和VEGF基因和蛋白水平的表達變化。結果:LPEE可顯著抑制HepG2和QGY-7703細胞穿過transwell小室和matrigel基質膠的細胞數(P0.05);50和100μg·mL~(-1) LPEE作用組與0組相比,可明顯降低VEGF mRNA和蛋白水平的表達(P0.05),而促進E-cadherin mRNA和蛋白水平的表達(P0.05)。結論:LPEE可抑制肝癌細胞的侵襲和遷移能力,其機制可能與抑制VEGF表達和促進E-cadherin表達有關。
[Abstract]:Objective: to investigate the effect of ethanol extract (Litsea pungens ethanol extract,LPEE on invasion and metastasis of hepatocellular carcinoma (HCC) cells and its related mechanism. Methods: IC50;transwell migration test and matrigel invasion test of LPEE on hepatocellular carcinoma cell lines HepG2 and QGY-7703 were detected by MTT assay and matrigel invasion test to detect the effect of LPEE on the migration and invasion of HepG2 and QGY-7703 cells. QRT-PCR and Western blot were used to detect the expression of E-cadherin and VEGF genes and proteins in each group by LPEE. Results: LPEE could significantly inhibit the number of HepG2 and QGY-7703 cells passing through transwell chamber and matrigel matrix gel (P 0.05). Compared with 0 group, 50 渭 g 路mL ~ (- 1) LPEE group significantly decreased the expression of VEGF mRNA and protein (P 0.05), but promoted the expression of E-cadherin mRNA and protein level (P 0.05). Conclusion: LPEE can inhibit the invasion and migration of HCC cells, and its mechanism may be related to the inhibition of VEGF expression and the promotion of E-cadherin expression.
