白細(xì)胞介素-17與肝癌家族聚集性的關(guān)系

【摘要】:目的研究白細(xì)胞介素-17在廣西肝癌高發(fā)地區(qū)肝癌高發(fā)家族成員與無(wú)癌家族成員血清中的表達(dá)情況,探討IL-17的表達(dá)水平與廣西肝癌家族聚集性的相關(guān)性。方法在廣西肝癌高發(fā)地區(qū)配對(duì)選取112對(duì)肝癌高發(fā)家族和無(wú)癌家族成員,共224例,高發(fā)家族組有男65例,女47例,年齡(27.1±21.2)歲,無(wú)癌家族組有男65例,女47例,年齡(26.8±17.7)歲。112例高發(fā)家族成員中根據(jù)親緣關(guān)系的不同,分一級(jí)親屬組、二級(jí)親屬組、三級(jí)親屬組、四級(jí)及以上親屬組,其中一級(jí)親屬62例,二級(jí)親屬11例,三級(jí)親屬25例,四級(jí)及以上親屬14例;根據(jù)高發(fā)家族組中患肝癌例數(shù)的不同,分2~3例組,4例及以上兩組,其中2~3例組70例,4例及以上組42例。以高發(fā)家族成員為實(shí)驗(yàn)組,無(wú)癌家族成員為對(duì)照組,采用酶聯(lián)免疫吸附(Enzyme-Linked Immunosorbent Assay,ELISA)法檢測(cè)兩組研究對(duì)象外周血清IL-17的表達(dá)水平。結(jié)果(1)高發(fā)家族組、無(wú)癌家族組IL-17的總體表達(dá)水平分別為(71.13±30.22)pg/ml、(19.01±14.46)pg/ml,高發(fā)家族成員IL-17表達(dá)水平顯著高于無(wú)癌家族成員,差異有統(tǒng)計(jì)學(xué)意義(t=16.464,P=0.000);進(jìn)一步分層分析:高發(fā)家族組男性、女性、年齡≤25歲、年齡25歲、HBs Ag陽(yáng)性、HBs Ag陰性、壯族、瑤族成員IL-17的平均濃度分別為(71.16±28.43)pg/ml、(71.08±32.86)pg/ml、(69.32±29.53)pg/ml、(73.73±31.33)pg/ml、(70.14±20.11)pg/ml、(71.82±35.76)pg/ml、(72.38±33.65)pg/ml、(67.99±19.25)pg/ml,無(wú)癌家族組男性、女性、年齡≤25歲、年齡25歲、HBs Ag陽(yáng)性、HBs Ag陰性、壯族、瑤族成員IL-17的平均濃度分別為(17.36±14.45)pg/ml、(21.30±14.30)pg/ml、(17.19±13.33)pg/ml、(21.63±15.71)pg/ml、(18.48±13.99)pg/ml、(19.39±14.87)pg/ml、(19.25±15.09)pg/ml、(18.41±12.93)pg/ml,兩組比較,t值分別為13.603、9.524、13.072、10.081、14.307、10.999、12.886、12.094,所有的P值均0.05,高發(fā)家族組不同人口學(xué)特征IL-17表達(dá)水平都高于無(wú)癌家族組,差異有統(tǒng)計(jì)學(xué)意義。(2)高發(fā)家族組中不同親緣系數(shù)的成員之間IL-17表達(dá)水平差異無(wú)統(tǒng)計(jì)學(xué)意義(Z=4.831,P=0.185)。(3)高發(fā)家族組中隨著患肝癌例數(shù)增加,IL-17表達(dá)水平下降,但差異無(wú)統(tǒng)計(jì)學(xué)意義(t=0.580,P=0.563)。結(jié)論肝癌高發(fā)家族成員IL-17表達(dá)水平升高與肝癌家族聚集現(xiàn)象存在相關(guān)性。
[Abstract]:Objective to study the expression of interleukin-17 (IL-17) in the serum of high incidence family members and non-cancerous family members of hepatocellular carcinoma (HCC) in Guangxi, and to explore the correlation between the expression of IL-17 and the aggregation of HCC family in Guangxi. Methods A total of 224 members of 112 high incidence and no cancer families in Guangxi were selected. There were 65 males and 47 females, aged (27.1 鹵21.2) years. There were 65 males and 47 females in the cancer free family group. Age (26.8 鹵17.7). 112 family members with high incidence were divided into first-degree relatives group, second-degree relatives group, third-degree relatives group, and four-grade relatives group, among which 62 cases were first-degree relatives and 11 cases second-degree relatives. 25 cases of third-degree relatives, 14 cases of four-grade and above relatives; According to the number of HCC cases in the high incidence family group, we divided them into 2 groups: 3 cases, 4 cases and more, including 70 cases in 23 cases, 42 cases in 4 cases and above. The expression of IL-17 in peripheral blood of the two groups was detected by enzyme linked immunosorbent assay (Enzyme-Linked Immunosorbent Assay,ELISA) with high incidence family members as experimental group and non cancerous family members as control group. Results (1) the total expression level of IL-17 was (71.13 鹵30.22) pg/ml, (19.01 鹵14.46) pg/ml, and (19.01 鹵14.46) pg/ml, in high incidence family and cancer free family respectively.
