【摘要】：目的檢測UCH37在結(jié)腸癌中的表達(dá),并分析其與患者的臨床病理特征的相關(guān)性,及其對結(jié)腸癌細(xì)胞增殖和凋亡的影響及分子機(jī)制。方法利用免疫組化檢測結(jié)腸癌組織中UCH37蛋白的表達(dá)情況,分析UCH37與結(jié)腸癌患者臨床病理特征的相關(guān)性,qPCR、CCK-8、流式細(xì)胞術(shù)和免疫共沉淀檢測UCH37對結(jié)腸癌細(xì)胞增殖、凋亡及MIF的去泛素化水平的影響。結(jié)果 UCH37蛋白在結(jié)腸癌組織中的陽性率高于癌旁組織,其表達(dá)水平與患者TNM分期及核分化水平呈正相關(guān);結(jié)腸癌細(xì)胞中UCH37的表達(dá)水平顯著高于正常結(jié)腸內(nèi)皮細(xì)胞;轉(zhuǎn)染UCH37 siRNA組細(xì)胞增殖能力低于轉(zhuǎn)染NC組,而凋亡細(xì)胞比例高于轉(zhuǎn)染NC組;感染UCH37慢病毒組增殖能力高于感染空載體組,而凋亡細(xì)胞比例低于感染空載體組。其機(jī)制為UCH37在結(jié)腸癌細(xì)胞中去泛素化MIF。結(jié)論 UCH37在結(jié)腸癌中表達(dá)升高,并通過去泛素化MIF促進(jìn)結(jié)腸癌細(xì)胞增殖。
[Abstract]:Objective to detect the expression of UCH37 in colon cancer and analyze its correlation with the clinicopathological features of colon cancer and its effect on the proliferation and apoptosis of colon cancer cells and its molecular mechanism. Methods the expression of UCH37 protein in colon cancer tissues was detected by immunohistochemistry, and the correlation between UCH37 and clinicopathological features of colon cancer patients was analyzed. QPCR,CCK-8, flow cytometry and co-immunoprecipitation were used to detect the proliferation of colon cancer cells by UCH37. Apoptosis and the effect of MIF deoxyuridine level. Results the positive rate of UCH37 protein in colon cancer tissue was higher than that in paracancerous tissue, and its expression level was positively correlated with TNM stage and nuclear differentiation level, and the expression level of UCH37 in colon cancer cell was significantly higher than that in normal colon endothelial cell. The proliferative ability of transfected UCH37 siRNA group was lower than that of transfected NC group, while the proportion of apoptotic cells was higher in infected UCH37 lentivirus group than that in infected empty vector group, while the proportion of apoptotic cells in infected UCH37 lentivirus group was lower than that in empty vector group. The mechanism is that UCH37 deubiquitin MIF. in colon cancer cells. Conclusion the expression of UCH37 in colon cancer is increased and the proliferation of colon cancer cells is promoted by MIF.
