白藜蘆醇對胃癌細(xì)胞及其表達(dá)DNA甲基轉(zhuǎn)移酶和組蛋白脫乙酰化酶的影響及意義
[Abstract]:Aim: to investigate the effect and significance of resveratrol (resveratrol,Res) on the expression of DNA methyltransferase (DNA methyltransferase,DNMTs) and histone deacetylase (Histone deacetylase,HDACs) in AGS gastric cancer cells. Methods: the cultured gastric cancer cells were cultured on the Res medium with 25 渭 mol/L,50 渭 mol/L,100 渭 mol/L for 24 hours. The apoptosis and cycle arrest of AGS were measured by flow cytometry. The expression of DNMT1,DNMT3a,DNMT3b,HDAC2,HDAC11 was detected by immunocytochemistry. Results: 24 hours after culture, the apoptosis of the control group, 25 渭 mol/L group, 50 渭 mol/L group and 100 渭 mol/L group were measured by flow cytometry (1.48 鹵0.151.77 鹵0.260.26 鹵26.59 鹵0.43) respectively. The G1 phase of cell cycle was measured by flow cytometry (44.47 鹵0.41U 52.15 鹵0.34U 55.22 鹵0.32U 71.48 鹵0.40F 2807.43p0.01). The gray values of DNMTs and HDACs by immunocytochemistry were DNMT1 (191.70 鹵0.60197.37 鹵0.73201.88 鹵0.29207.97 鹵0.81FU 347.21 p0.01) and DNMT3a (187.64 鹵0.96197.21 鹵0.63203.35 鹵0.74208.17 鹵0.30F481.49p0.01), respectively. DNMT3b (191.69 鹵0.65196.36 鹵0.44201.04 鹵0.37205.69 鹵0.46FU 452.17p0.01), HDAC2 (184.58 鹵1.01193.32 鹵0.57200.60 鹵0.84205.12 鹵0.53FU 414.68p0.01). HDAC11 (189.94 鹵0.70194.14 鹵0.99201.71 鹵0.73207.46 鹵0.777). Conclusion: 1. In a certain concentration range, Res could inhibit the production of DNMTs and HDACs, accelerate the apoptosis of gastric cancer cells, and block the cell cycle in G1 phase.
